摘 要:目的研究加压素(vasopressin,VP)对大鼠视前区-下丘脑前部(preoptic-anterior hypothalamus,PO/AH)神经元自发放电的紧张性调节作用。方法制备大鼠下丘脑片,通过红外微分干涉相差技术和脑片膜片钳技术,采用全细胞模式记录PO/AH神经元放电,鉴定温度敏感类型。脑片灌流给予1μmol/L VP或VP V1a受体阻断剂,观察对热敏神经元、冷敏神经元和温度不敏感神经元自发放电的影响。结果与自身对照比较,外源性VP引起热敏神经元放电频率升高(P<0.01),冷敏神经元放电频率下降(P<0.05),而温度不敏感神经元放电有兴奋,也有抑制作用(均P<0.01)。阻断V1a受体,产生的作用相反(均P<0.05)。结论 VP通过V1a受体对大鼠PO/AH热敏神经元、冷敏神经元和温度不敏感神经元自发放电均产生紧张性调节作用。Objective To investigate the tonic regulatory role of vasopressin (VP) in spontaneous firing of neurons in the preoptic-anterior hypothalamus (PO/AH) of rats. Methods Rat PO/AH tissue slices were prepared. The firing activity of PO/ AH neurons was recorded and the thermosensitivity of neurons identified by whole-cell patch clamp recording and infrared dif- ferential interference contrast technology. The spontaneous firing activity of warm-sensitive neurons, cold-sensitive neurons, and temperature-insensitive neurons was observed when PO/AH slices were perfused with 1 μmol/L VP or V1a vasopressin receptor antagonist. Results The exogenous VP increased the spontaneous firing activity of warm-sensitive neurons (P〈0.01) and de creased that of cold-sensitive neurons (P〈0.05). Some of temperature-insensitive neurons were excited by the exogenous VP while others were inhibited (P〈0.01). Blocking Via receptor induced the opposite effect (P〈0.05). Conclusion The tonic regulatory role of VP in spontaneous firing of rat PO/AH warm-sensitive neurons, cold-sensitive neurons, and temperature insensitive neurons is induced by the V1a vasopressin receptor.
