和胃煎剂对实验性萎缩性胃炎小鼠胃黏膜再生的影响

摘　　要：目的通过观察胃黏膜细胞再生情况,探讨和胃煎剂治疗慢性萎缩性胃炎的机制。方法 45只5周龄雄性BALB/c小鼠随机分为正常组、萎缩性胃炎组以及干预组,以水杨酸钠法复制慢性萎缩性胃炎小鼠模型,HE及免疫组化法检测胃黏膜陷凹细胞,壁细胞,增殖细胞的变化;半定量实时荧光PCR检测小鼠胃黏膜msi-1 mRNA表达。结果萎缩性胃炎小鼠壁细胞,陷凹细胞减少显著低于正常组与干预组(P<0.01),PCNA阳性表达增加显著高于正常组与干预组,msi-1 mRNA表达显著高于干预组和与正常组(P<0.01);干预组与正常组之间比较差异无统计学意义(P>0.05)。结论和胃煎剂对水杨酸钠法制备的小鼠慢性萎缩性胃炎具有保护作用,其机制可能与诱导壁细胞分化有关。Objective To explore the effect of Hewei Decoction （decoction for regulating the stomach） on the mucosal cell regeneration in the mouse model of chronic atrophic gastritis （CAG）. Methods Forty-five BALB/c mice were randomly divided into three groups： normal control, CAG and Hewei Decoction intervention groups. The mouse models of CAG were prepared by oral intake of sodium salicylate. During the same period, Hewei Decoction was administrated in the intervention group. Hematoxylin and eosin （HE） staining and immunohistochemistry were applied to observe the changes of mueosal pit cells, parietal cells and proliferative cells. Real-time quantitative polymerase chain reaction （PCR）was performed to detect the expression of msi-1 mRNA in mucosal specimens. Results Compared with those in the normal control and Hewei-Decoction intervention groups, pit cells and parietal cells reduced significantly, and the expression of PCNA positive cells and msi-1 mRNA level were significantly increased in CAG group （P 〈 0. 01 ）. There were no significant differences in the above-mentioned parameters between Hewei-Decoction intervention group and normal control group（P 〉 0. 05）. Conclusions The Hewei Decoction can relieve chronic atrophic gastritis induced by sodium salicylate in mouse, which may be related to the differentiation of oxyntic cell lineage.
