Chiral Separation by Dynamically Dual-coating Capillary Electrochromatography Using Cationic Surfactant and Anionic Chiral Selector as Pseudo-stationary Phases

摘  要:建立了以十六烷基三甲基溴化铵或1 ,5_二甲基_1 ,5_二氮杂十一烷亚甲基聚N_甲溴化物为阳离子表面活性剂 ,并以磺丁基 β_环糊精为手性选择剂的双动态吸附毛细管电色谱。以碱性的丙比胺和酸性的华法林作为拆分对象 ,考察了双动态吸附毛细管电色谱的手性分离行为 ,以及动态吸附柱的重复性。在双动态吸附毛细管电色谱条件下 ,丙比胺和华法林的手性分离度较大 ,丙比胺的分离度可达3.21 ,丙比胺连续进样10次 ,迁移时间的相对标准偏差小于1.0 %。A novel capillary electrochromatography(CEC) with cationic surfactant hexadimethrine bromide(HDB) or cetyltrimethylammonium bromide(CTAB) and anionic chiral selector of sulfonyl butyl ether _ β _ cyclodextrin(SBE _ β _ CD) as pseudo _ stationary phases was established. The cationic surfactant firstly dynamically coated onto the negative charged surface of silica column due to electrostatic interaction. Then anionic SBE _ β _ CD was dynamically adsorbed as the second coating onto the cationic surfactant coating. The effect of concentrations of CTAB, SBE _ β _ CD and buffer pH on chiral separation was investigated. Basic disopyramide and acidic warfarin were used as tested analytes. The capability of chiral separation of this dynamically dual _ coating CEC proved to be stronger than that of CD-CZE. The resolution of 3.21 for disopyramide was achieved, and the RSD of migration time was less than 1.0%(n=10).
