摘 要:提出了测定人体尿液中芦氟沙星的Eu3 +敏化荧光法 ,研究了芦氟沙星在HAc介质中被H2O2 氧化的反应机理。在近中性HAc -NaAc缓冲溶液中芦氟沙星的氧化产物与Eu3 +、EDTA形成三元络合物 ,产生Eu3 +的特征荧光 (λem 617nm、λex 352nm) ,其荧光强度与芦氟沙星的浓度成线性关系。尿液标准曲线线性范围5.0×10-8~2.5×10-6 mol/L ,检出限1.5×10-8 mol/L。方法简单、快速、灵敏。Rufloxacin can be oxidized by H2O2 in HAc solution at 100 ℃, and its fluorescence is lost. A ternary complex of the oxidation product with EDTA and Eu3+ can be formed in the HAc-NaAc buffer solution. The complex has characteristic fluorescence of Eu3+ at an emission wavelength of 617 nm(λex=352 nm). The linear relationship between fluorescence intensity and concentration of rufloxacin in the range of 5.0×10-8~2.5×10-6 mol/L was found. A method for the determination of rufloxacin in human urine was establised by Eu3+ sensitized fluorescence analysis in view of the above. Under the selected optimum conditions, the detection limit of the method is 1.5×10-8 mol/L. The method is simple, rapid and sensitive. The mechanism of the oxidation reaction of rufloxacin in HAc solution was also discussed according to the changes of UV, fluorescence and infrared spectra.
