摘  要:目的:探讨五味消毒饮水煎液对乳腺上皮细胞炎症模型的作用及对大肠杆菌感染小鼠的保护作用机制。方法:将96孔板细胞分为对照组、模型组、治疗组和中药组。将五味消毒饮水煎液按终浓度0.2 g/mL、0.4 g/mL和0.8 g/mL添加到乳腺上皮细胞炎症模型中,观察细胞形态,用MTT法检测各组细胞OD值;将24只小鼠随机分为对照组、模型组和治疗组,每组8只,将1 g/mL的五味消毒饮水煎液腹腔注射于炎症模型小鼠,观察小鼠临床症状,以ELISA法检测血清白细胞介素-10(IL-10)的表达水平。结果:五味消毒饮水煎液0.8 g/mL使乳腺上皮细胞固缩、脱壁;0.2 g/mL对细胞炎症模型有显著的保护作用,细胞形态与对照组相同。五味消毒饮水煎液(1 g/mL)对小鼠大肠杆菌感染有保护作用,72 h内,对照组饮水、摄食均衡,精神状态良好,零死亡;模型组饮水、摄食减少,精神萎靡不振,死亡7只,治疗组死亡1只。与对照组比较,模型组小鼠血清IL-10显著升高(P<0.05);治疗组IL-10浓度较模型组低,与对照组比较无显著差异,五味消毒饮有降低炎症小鼠血清IL-10浓度的趋势。结论:五味消毒饮水煎液0.2 g/mL对细胞炎症模型有显著的保护作用,1 g/mL对小鼠大肠杆菌感染有保护作用,且能降低炎症小鼠血清IL-10浓度,促进小鼠免疫机能发挥作用。Objective: To explore the mechanism of Chinese herbal preparation Wuwei Xiaoduyin against mouse infection by Escherichia coli and its effect on Holstein-bovine mammary epithelial cells models of inflammatory response.Methods: Different concentration of Wuwei Xiaoduyin extract was added on inflammation model. The morphology change of the cells was observed and the light absorption value was determined using MTT method. Then clinical status was observed and ELISA was employed to determine the level of Interleukin-10(IL-10) in serum. Results: Almost all cells treated by 0.8 g/mL of Wuwei Xiaoduyin were broken, while 0.2 g/mL of the preparation could protect cells models of inflammatory response, which was similar to the cells in control group. It also had protective effects on mouse infection model. Compared with control group, model group experienced a remarkable rise in expression of IL-10(P〈0.05) in serum; Expression of IL-10 in Wuwei Xiaoduyin treated group was increased compared to control group. Conclusion:Wuwei Xiaoduyin has protective effects on both cells models of inflammatory response(0.2 g/mL) and mouse infection model(1 g/mL). It could enhance the immune function by decreasing the expression of IL-10 in serum of infected mice.
