摘  要:为了优化大孔树脂提取1012-4菌株发酵液中抗耐甲氧西林金黄色葡萄球菌活性物质的工艺,通过多种型号比较,确定大孔树脂型号,然后基于单因素实验探讨最优工艺。实验结果表明:最优树脂型号为NKA-2,其吸附量为5%;洗脱溶剂及浓度为15%的乙醇溶液,洗脱体积为6 BV,吸附流速为2 mL/min,解吸附流速为1 mL/min。该方法环保、节能,可操作性强,可用于工业化生产。To optimize the macroporous resin extraction process of active substance which has the ac- tivity of anti-MRSA from microbial fermentation broth, the most effective resin and the operation con- ditions were selected by single factor experiment. The optimal adsorption resin for extracted active substance is NKA-2. The main technical parameters are as follows: adsorption volume is 5%, the 15% ethanol as eluent solvent, washing volume is 6 BV, adsorption velocity is 2 mL/min, desorption velocity is 1 mL/min. The method has the characteristics of environmental protection, energy saving, and easy operation which can be used for industrial production.
