摘 要:目的:观察舒郁胶囊对血管性抑郁模型大鼠脑组织5-羟色胺1A受体(5-HT1AR)、多巴胺2受体(D2R)、α肾上腺素2A受体(α2AR)和载脂蛋白E4(Apo E4)表达的影响。方法:除正常组外其他各组大鼠分别im给予维生素D3(VD3)30万U·kg-1,连续3 d,第4天起给予高脂饲养9周,继续高脂的同时给予慢性不可预见性温和应激刺激(CUMS)3周,建立大鼠血管性抑郁模型。舒郁胶囊低、中、高剂量组大鼠均ig给予舒郁胶囊533,1 066,2 132 mg·kg-1,氟西汀组给予盐酸氟西汀1.3 mg·kg-1,正常组及模型组大鼠ig等体积蒸馏水,各组均为1次/d,疗程共21 d。以敞箱实验,糖水偏嗜实验进行行为学评分,采用免疫组织化学法检测脑内5-HT1AR,D2R,α2AR的含量变化,采用酶联免疫法测定Apo E4的含量变化。结果:与正常组比较,模型组大鼠脑内的5-HT1AR,D2R,α2AR受体和Apo E4的含量表达显著增加(P<0.01);舒郁胶囊低、中、高剂量组和氟西汀组与模型组比较均能降低大鼠脑内5-HT1AR,D2R,α2AR受体及降低Apo E4含量(P<0.05,P<0.01),舒郁胶囊3剂量组和氟西汀组之间比较无明显差异。结论:舒郁胶囊可能具有抗血管性抑郁作用,其作用机制可能与降低大鼠脑内5-HT1A R,D2R,α2AR及Apo E4表达有关。Objective: To observe the effect of shuyu capsule on the expression of serotonin 1A receptor (5-HTIAR), dopamine D2 receptor (DzR), α 2A adrenergic receptor ( α2AR) and apolipoprotein E4 (Apo E4) in brain tissue of rats with vascular depression model. Method: Except the normal group rats were given Vitamin D3 (VD3) 300 000 U·kg^-1 byintramuscular injection, for 3 d, at fourth day high-fat feeding was caried out, lasted for 9 weeks, meanwhile chronic unpredictable mild stress (CUMS) was performed for 3 weeks to establish the rat model of vascular depression. Shuyu capsule low dose (533 mg·kg^-1), middle dose (1 066 mg·kg^-1 ), high dose (2 132 mg·kg^-1 ) and fluoxetine ( 1.3 mg ·kg^-1 ) were administered intragastrically group were given fluoxetine hydrochloride, the normal group and the model group rats were administrated distilled water, the treatment lasted 21 days. In the open field test, sucrose preference test was scored, immunohistochemical method was used to detect the changes of content of 5-HT1AR in brain, D2R, ol 2^R, Apo E4 content determinationby ELISA method. Result: The 5-HT1AR, D2R, α2AR receptor and Apo E4 expression increased significantly inthe model group (P 〈0. O1 ). Shuyu capsule low, middle, high dose groups and fluoxetine group, compared withthe model group, could reduce the 5-HT1AR and D2R,α2AR receptor and decreased the content of Apo E4 (P 〈O. 05, P 〈0.01), Conclusion: Shuyu capsule may have anti-vascular depression effect, its mechanism may berelated to the decrease of 5-HT1AR, D2R, α2AR and expression of Apo E4.
