摘  要:目的研究桑枝水提物中降糖活性部位和化学成分。方法以α-葡萄糖苷酶为靶点,酶标仪检测分析,结合各种柱层析法对具有最强抑制作用的桑枝活性部位进行分离,确定活性成分;体内实验以正常雄性ICR及实验性糖尿病小鼠为实验对象,以阿卡波糖为阳性对照,采取鼠尾尖血测血糖值的方法进行糖耐量实验。结果桑枝水提物用AB-8型大孔吸附树脂粗分,穿透液(未被吸附)部位及硅胶柱乙酸乙酯∶无水乙醇(1∶4)洗脱部位对α-葡萄糖苷酶的抑制活性最强,其抑制率分别为36.97%和59.93%;分离纯化得到的化合物(1-脱氧野尻霉素)对α-葡萄糖苷酶的抑制效果达到IC50为0.004mg/L,其对正常小鼠和糖尿病小鼠均能很好的降低其餐后血糖水平。结论桑树水提物有抑制α-葡萄糖苷酶活性及降低小鼠餐后血糖水平的作用。Objective To investigate the chemical constituents of water extract from the Ramulus mori for hypoglycemic. Methods Set alpha - glycosidase enzymes as target, analysis by ELIASA , separation of strongest inhibitory combining all sorts of column chromatography , determine the active ingredients ; set male ICR normal and experimental - diabetic mice as experimental object in vivo , acarbose as positive control, the experiment of glucose tolerance by the determination of blood glucose levels in mice tail tip. Results The components has not been absorbed by AB -8 macroporous resin and the components which elution down by ethyl acetate : anhydrous ethanol ( 1 : 4 ) on silica gel column hold the best inhibitory activity for alpha - glucosidase, the inhibition rate were 36.97% and 59.93% respectively. The compound (DNJ) which was separated from active site indicated a good inhibitory activity for alpha - glueosidase,its IC50 was only 0. 004 mg/L and it can well reduce the PBS levels on both normal mice and dia- betic mice. Conclusion Ramulus mori extract has the inhibition of α- glycosidase enzymes and reduces the PBS leves of mice.
