摘 要:目的优选复方虎杖颗粒的提取纯化工艺条件。方法以虎杖苷含量和水浸出物量为综合评价指标,采用L9(34)正交试验考察加水量、煎煮次数、煎煮时间3个因素对提取工艺的影响;以除杂率和虎杖苷转移率为考察指标,采用单因素试验考察醇浓度和清膏相对密度对醇沉工艺的影响。结果复方虎杖颗粒的最佳提取工艺为依次加12,10,10倍量水煎煮3次,每次0.5 h;最佳纯化工艺条件为将提取液浓缩至相对密度1.13(70℃)的清膏,加乙醇至含醇量为80%,静置24 h。结论优化所得提取纯化工艺可最大限度的保留有效成分,并达到了较好的去杂效果,为确定虎杖苷的提取纯化工艺提供了实验依据。Objective To optimize water extraction and alcohol precipitation process for compound recipe Hu - zhang granule. Methods The amount of water extracts and polydatin were used as comprehensive evaluation indexes, orthogonal test was used to optimize extraction technology with the amount of water, decocting times and decocting time as factors. Single - factor test was used to optimize alcohol precipitation process with the concentration of alcohol precipitation and relative density of extract were chosen as factors. Results Optimum water extraction process was as follows: decocted 3 times with 12,10,10 times the amount water,0.5 h each time; optimum alcohol precipitation process was: concentrated extraction liquid to its relative density 1.13 (70 ℃ ), added alcohol until the concentration of ethanol at 80%, stilled 24 h. Conclusion These optimized of water extraction and alcohol precipitation process could ensure effective ingredient saved to the maximum and achieve good impurity effect. The experi- ment result can provide basic and instruction data for the ascertainment of water extraction technology of polydatin.
