摘　　要：目的建立一种适合于广地龙干燥药材DNA的提取方法。方法参考陈旧皮张标本的DNA提取方法,并对样品预处理、酶解条件、沉淀时间等对影响DNA质量的重要因素,进行了筛选和优化。结果按优选的方法提取的总DNA的含量和纯度完全符合引物扩增分析的要求。结论该提取方法不仅解决了地龙类动物干燥药材DNA提取的难题,而且操作简便实用、DNA提取效率高,所得DNA的质量也满足后期DNA分子鉴定。Objective To improve and optimize the extraction process of DNA from medicinal Guang Dilong. Methods A new feasible DNA extraction method was available by modifying and optimizing the extraction method of DNA from old or dried skins. Results By modifying the extraction method of DNA from old or dried skins, the content and purity of total DNA were all up to the standards for PCR analysis. Conclusion A simple and feasible extraction process of DNA from medicinal Guang Dilong was created and the content and purity of the DNA extracted in this method can meet the requirements for DNA molecular identification.
