牛补体C6基因SNPs与奶牛乳腺炎的关联性

摘　　要：以469头中国荷斯坦奶牛为样本,利用DNA测序及多态性聚合酶链反应(PCR-RFLP)技术等方法检测牛C6基因中单核苷酸多态性(SNP)位点。经序列比对发现4个单核苷酸多态性位点(SNPs):第2外显子上g.+2235G>A(G突变为A),第8外显子上g.+32881C>A(C突变为A)和g.+32925C>T(C突变为T),第17外显子上g.+69060T>C(T突变为C)。经SAS软件统计分析发现:g.+32881C>A突变的群体,CA基因型的个体体细胞评分显著高于CC基因型的个体(p<0.05),且其产奶量显著低于CC个体(p<0.05);发生g.+69060T>C突变的个体,CC基因型的个体产奶量极显著高于TT基因型个体(p<0.01),同时其乳蛋白率显著高于TT基因型个体(p<0.05)。The complement system is the humoral immune effector of the innate immune mechanisms regpulatory system which is present in the plasma and the on the cell surface proteins. In our study, we have detected the single nucleotide polymorphism sites （SNPs） of the C6 gene by using the method of DNA sequencing and PCR-RFLP in 469 Holstein eattles. We found four SNPs：g.＋ 2235 G 〉 A mutation in exon 2 ,g.＋ 32881 C 〉 A and g.＋ 32925 C 〉 T mutations in exon 8, g.＋ 69060 T 〉 C mutation in 17 exons. Studies have shown ： the somatic cell count of CA genotype was significantly higher than CC genotype （p 〈 0.05 ） in groups with g.＋ 32281 C 〉 A mutation, and the milk yield was significantly higher than the AG individuals （p 〈 0.05 ） ;in the groups with g.＋ 69060 T 〉 C muta- tion, the individual milk yield of the CC genotype was significantly higher than TT genotype （p 〈 0.01 ） and the milk protein was signifi- cantly higher than TT genotype （p 〈0.05 ）.
