摘  要:以大花蕙兰无菌类原球茎为材料,分别研究了不同生长物质对类原球茎增殖与分化的影响,以及不同浓度海藻酸钠包埋与不同培养基质对于人工种子萌发的影响。结果表明,适宜类原球茎增殖的培养基是1/2 MS+2,4-D2.0 mg/L+NAA 0.2 mg/L;培养基中添加终浓度为1.0 mg/L的活性炭,可有效降低类原球茎褐化的发生;适于类原球茎分化成苗的培养基是MS+0.5 mg/L PP333;用2%海藻酸钠包埋的人工种子在添加了2,4-D 2 mg/L的MS培养基质中萌发率较高。Protocorm-like bodies (PLBs) of Cymbidium hybridum were used as materials to study effects of different growth substances on proliferation and differentiation of PLBs as well as different concentrations of sodium alginate and different culture media on germination of artificial seeds.The results showed that the suitable medium for the proliferation of PLBs was 1/2 MS + 2,4-D 2.0 mg/L + NAA 0.2 mg/L; the medium supplemented with 1.0 mg/L of activated carbon could effectively reduce the browning of PLBs;the suitable medium for protocorm differentiate into seedling was MS + 0.5 mg/L PP333 ; The germination rate of artificial seeds entrapped with 2% sodium alginate on MS culture medium added with 2,4-D 2 mg/L was higher.
