摘 要:目的研究三种常见的有机紫外防晒剂与牛血清白蛋白(BSA)相互作用的荧光光谱特征。方法利用荧光光谱法,观察扫描温度分别为300、305、310 K时牛血清白蛋白与不同浓度的有机紫外防晒剂相互作用的荧光光谱,判断荧光猝灭类型并分析结合能力和结合模式。结果三种有机紫外防晒剂均对BSA有荧光猝灭作用,猝灭类型为静态猝灭;均与BSA有较强的结合能力,结合能力大小为丁基甲氧基二苯甲酰基甲烷(BM-DBM)>二苯酮-3(BP-3)>3-(4-甲基苯亚甲基)-樟脑(4-MBC),且均与BSA可形成一个结合位点;BP-3和4-MBC与BSA之间的主要作用力类型为氢键和范德华力,BM-DBM与BSA之间的主要作用力类型为疏水作用力。结论 BP-3、4-MBC、BM-DBM均能与牛血清白蛋白发生结合作用。Objective To study the fluorescence spectral characteristics of the interaction between three common organic UV filters and bovine serum albumin(BSA). Methods The fluorescence spectrums of the interaction between BSA and UV filters of different concentrations was scanned under 300, 305, 310 K, respectively. In addition, the type of fluorescence quenching was determined and the binding affinity and binding mode were analyzed. Results The selected three organic UV filters had fluorescence quenching effects on BSA. The interaction process was further identified as static quenching. The binding affinity of three UV filters was in the order of BP-3〉4-MBC〉BM-DBM. Each of them could form one binding site with BSA. The reaction type between BP-3,4-MBC and BSA was given priority to hydrogen bond and Van der Waals force. However,it was mainly hydrophobic force between BM-DBM and BSA. Conclusion BP-3,4-MBC and BM-DBM all can combine with bovine serum albumin.
