摘 要:目的:按传统药材外观分级指标对丹参饮片分级,用HPLC法测定净丹参饮片中丹酚酸B和丹参酮ⅡA的含量,建立主成分含量与丹参饮片分级标准相关性。方法:按传统药材分级标准,对丹参饮片分级。色谱柱采用依利特C18BP,丹酚酸B以甲醇-0.5%甲酸水溶液(40%~60%,60%~40%)15 min梯度洗脱,检测波长286 nm;丹参酮ⅡA以甲醇-水(87∶13)洗脱,检测波长为270 nm。结果:丹酚酸B在1.20~9.60μg·ml-1范围内线性良好(r2=0.999 3),回收率为99.76%(RSD为0.63%,n=5),饮片外观分级与丹酚酸B含量呈正相关性,各级别饮片中丹酚酸B含量有明显差异;丹参酮ⅡA在0.050 4~1.008μg·ml-1范围内线性良好(r2=0.999 2),回收率为98.38%(RSD为1.07%,n=5)。饮片外观分级与丹参酮ⅡA含量无相关性,不同产地、不同级别的净丹参饮片中丹参酮ⅡA含量有差异。结论:可将丹酚酸B含量作为丹参饮片分级的指标,其中优级含量大于7.4%。OBJECTIVE To determine the contents of salvianolic acid B and tanshinoneⅡin different grades of Salvia Miltiorrhiza slices,and to construct the relevance between different grades of slices of Radix Salviae Miltiorrhizae and contents of major ingredients.METHODS According to traditional classified criterion of Chinese traditional drug's appearance,the different grades of slices of Radix Salviae Miltiorrhizae were classified,the contents of salvianolic acid B and tanshinoneⅡin different grades of Salvia Miltiorrhiza slices were determined by using HPLC.C18BP chromatographic column with methanol and0.5%formic acid(40%-60%∶60%-40%)15 min gradient elution was used,and the detection wavelength was 286 nm for salvianolic acid B;methanol and water(87∶13)was used as mobile phase,and the detection wavelength was 270 nm for tanshinoneⅡA.RESULTS Salvianolic acids B in1.20-9.60μg·ml-1 had a good linear range(r2=0.999 3),the average recovery was 99.76%(RSD was 0.63%,n=5).While tanshinoneⅡA in 0.050 4-1.008μg·ml-1 had a good linear range(r2=0.999 2),the average recovery was 98.38%(RSD=1.07%,n=5).It was shown that different grades of slices of Radix Salviae Miltiorrhizae had distinct differences in their content of salvianolic acid B and tanshinoneⅡA.There was better relevance between the contents of salvianolic acid B and grades of appearance,but there was irrelevance between the contents of tanshinoneⅡA and grades of appearance.CONCLUSION The content of salvianolic acid B can be used as the index to classify the grades of slices of Radix Salviae Miltiorrhizae,the contents of salvianolic acid B should be over 7.4%as superfine grade.
