摘 要:目的 观察依托咪酯(etomidate,Eto)和氯胺酮(ketamine,Ket或K)合用对小鼠学习记忆的影响.方法 实验共用昆明种小鼠80只:避暗法(step-through text)和跳台法(step-down text)各40只,再分为4组(n=10):生理盐水(NS)组、依托咪酯(Eto)组、氯胺酮(Ket)组和两药合用(Eto+ K)组,分别观察各组潜伏期和错误次数.结果 Eto+K组用药24 h后避暗潜伏期(29.1±15.7)s短于NS组[(193±112)s,P<0.01]、Eto组[(127±124)s,P<0.05]和Ket组[(197±133)s,P<0.01],错误次数(1.9±0.6)多于NS组(0.7±0.7,P<0.01)、Eto组(1.1±0.9,P<0.05)和Ket组(0.7±1.1,P<0.01);Eto+K组用药48 h后避暗法潜伏期(147±134)s短于NS组[(256±78.0)s,P<0.05]和Eto组[(252±79.1)s,P<0.05],错误次数差异无统计学意义(P>0.05);Eto+K组用药24 h后跳台法潜伏期(133±126)s短于NS组[(253±72.9)s,P<0.01]、Eto组[(235±72.0)s,P<0.05]和Ket组[(224±67.9),P<0.05],错误次数(1.3±1.1)多于NS组(0.4±0.5,P<0.05)和Eto组(0.5±0.5,P<0.01);Eto +K组用药48 h后各组潜伏期和错误次数的差异无统计学意义(P>0.05).结论 依托咪酯和氯胺酮合用后至少24 h内抑制小鼠学习记忆呈协同作用,但用药后48 h协同作用减弱,不会造成持久的学习记忆障碍。Objective To observe the effects of a combination of etomidate (Eto) and ketamine (Ket or K) on learning and memory in mice.Methods 80 Kunming mice were divided into two groups for step-through and step-down tests (40 in each).The mice in each group were subdivided into four groups (n =10):a normal saline (NS)group,an etomidate (Eto) group,a ketamine (Ket) group and a combined treatment (Eto + K) group.Then,the latency and error times of each group were observed.Results Twenty-four hours after injection,the latency in step-through test of the Eto + K group (29.1 ± 15.7) s were shorter than those of the NS group [(193 ± 112) s,P 〈 0.01)],the Eto group [(127 ± 124) s,P 〈 0.05],or the Ket group [(197 ± 133) s,P 〈 0.01].Meanwhile the Eto + K group presented more error times (1.9 ± 0.6) than the NS group (0.7 ± 0.7,P 〈 0.01),the Eto group (1.1 ± 0.9,P 〈0.05),and the Ket group (0.7 ± 1.1,P 〈0.01).Forty-eight hours after injection,the latency in step-through test of the Eto + K group (147 ± 134) s were remarkably shorter than those of the NS group [(256 ± 78.0) s,P 〈 0.05] or the Eto group [(252 ± 79.1) s,P 〈 0.05],while no statistical differences were found in error time among these groups (P 〉 0.05).Twenty-four hours after administration,the latency in step-down test of the Eto + K group (133 ± 126) s were shorter than those of the NS group [(253 ± 72.9) s,P 〈 0.01],the Eto group [(235 ± 72.0) s,P 〈 0.05],or the Ket group (224 ± 67.9).Meanwhile,more error times were observed in the Eto + K group (1.3 ± 1.1) than the NS group (0.4 ± 0.5,P 〈 0.05) and the Eto group (0.5 ± 0.5,P 〈 0.01).No statistical differences were seen in the step-down latency and error time of the Eto + K group 48 hours after administration (P 〉 0.05).Conclusion A combination of etomidate and ketamine produces synergistic effects on learning and memory damage in mice within 24
