摘 要:用离体大鼠胸主动脉血管环为研究对象,经生物信号采集系统采集并分析血管环张力变化,观察霉茶总黄酮、二氢杨梅素、杨梅素对静息血管的影响以及它们对去甲肾上腺素(NA)所致的缩血管反应的影响。研究发现霉茶总黄酮、二氢杨梅素、杨梅素均可浓度依赖性增加血管环静息张力;霉茶总黄酮能使NA缩血管的量-效曲线非平行右移并降低其效能(Emax);二氢杨梅素、二氢杨梅素与杨梅素的混合物有与霉茶总黄酮相似的效应;二氢杨梅素与杨梅素合用使NA收缩血管的Emax降低,明显强于二者单独使用;当二氢杨梅素与杨梅素浓度比为10∶1时,拮抗NA缩血管的效应最佳。结果显示霉茶总黄酮对静息血管有剂量依赖性收缩作用但能明显拮抗NA缩血管效应,二氢杨梅素与杨梅素对NA诱导血管收缩反应有协同增效作用且最佳协同比例为10∶1。To study the effect of total flavonoids of Ampelopsis megalophylla, dihydromyricetin and myricetin on thoracic aorta rings of rats, a biological signal collection and analytical system was applied to observe the tension of resting thoracic aorta rings and the dose-effect curve of the contractile response induced by noradrenaline (NA). The results showed that A. megalophylla tlavonoids, dihydromyricetin and myricetin can increase the tension of resting thoracic aorta rings in a dose-depondent manner. The total flavonoids of A. megalophylla shifted the dose-effect curve of the contractile response induced by NA to the right and decreased their maximal efficiency (Emax). The similar effects were also observed in di- hydromyrieetin and the mixture of dihydromyricetin and myricetin, wherein the mixture lowered much more Emax than either of the two isllets applied individually. In particular, the mixture comprising of dlhydromyricetin and myricetin with concentration ratio of 10:1 showed better effect. In conclusion, flavonoids of A. megalophyUa induced dose-dependent contraction in isolated thoracic aorta rings in rats and can significantly noncompetitively antagonize NA induced contraction of rat aortic rings. In addition, dihydromyficetin and myricetin synergistically antagonize NA induced contraction of vascular rings with optimal ratio of 10: 1.
