摘  要:目的探讨中药大黄素对单侧输尿管梗阻大鼠肾组织中基质金属蛋白酶-9(MMP-9)表达的影响。方法 120只wister大鼠随机分成3组:假手术组、梗阻组及大黄素治疗组。制备单侧输尿管梗阻(UUO)模型。治疗组在UUO的基础上每天以大黄素40mg/kg灌胃,采用病理及免疫组化的方法动态观察各组大鼠术后1d、3d、7d、14d梗阻侧肾脏组织学变化和MMP-9在梗阻肾小管间质中表达情况。结果模型组MMP-9的表达先增高后降低,治疗组MMP-9的表达水平高于模型组(P<0.05)。结论中药大黄素可以通过提高MMP-9的表达,促进ECM的降解来发挥其延缓肾间质纤维化进程的作用。Objective To investigate the dynamic changes of MMP-9 expressions in renal interstitial flbrosis(RIF), and to explore the mechanism of RIF and the effect of emodin. Methods One hundred and twenty rats were randomly divided into 3 groups: the sham-operated group, the UUO group and the ernodin treated group. Rat in the DTG were administered intragastrically with emodin 40mg/kg daily after modeling. Ten rats of each group were killed respectively at day 1,3,7 and 14 days after UUO. Renal histopatholngical changes was observed by light microscope. The expression of MMP-9 were examined by HE staining and immunohistochemistry at each time point. Results In UUO rats, the express of MMP-9 firstly increased and then decreased;In DTG group, the express of MMP-9 incrased significantly compared to the same time in the UUO group. ConclusionThe up-regulation of MMP-9 involved in the pathogenesis of tubulointerstitial fibrosis. Emodin may suppress fibrosis development and can protect the function of renal.
