摘 要:目的探讨蓬子菜总黄酮(FGVL)抗急性早幼粒细胞白血病细胞株NB4的作用及诱导凋亡的分子机制。方法噻唑蓝法检测FGVL对细胞增殖的抑制作用;吖啶橙/溴乙锭荧光染色观察NB4细胞凋亡的形态学变化;DNA琼脂糖凝胶电泳检测细胞凋亡;逆转录聚合酶链式反应检测凋亡相关基因Bcl-2及Bax表达。结果 FGVL明显抑制NB4细胞增殖,FGVL50、100、200μg/mL组细胞增殖抑制率分别为(24 h:6.7%、7.4%、24.3%,48 h:2.3%、18%、45.9%,72h:3.7%、24%、51.7%);细胞形态学观察可见明显的细胞凋亡特征;并有典型的DNA"梯形"凋亡条带出现。FGVL下调Bcl-2 mRNA表达,上调Bax mRNA表达,呈剂量和时间依赖关系。结论 FGVL可抑制急性早幼粒细胞白血病NB4细胞的增殖并诱导细胞凋亡,其作用机制与调节Bcl-2/Bax表达有关。Objective To study anti-proliferation and apoptosis promotion effect of flavone from Galliu Verum L (FGVL) on acute promyelocytic leukemia ceils ( NB4 ) and its possible mechanisms. Methods The proliferation inhibi- tion of NB4 cells was detected with 3- (4,5) dimethylthiazol-2-yl-2,5-di-phenyltetrazolium bromide (MTT). DNA agarose electrophoresis was used to detect cell apotosis and acridine orange ethidium bromide(AO/EB) staining was used to observe morphological changes of NB4 cells. The expression of pro- and anti-apoptotic genes Bax and Bcl-2 were examined with reverse transcription polymerase chain reaction. Results FGVL at the doses of 50,100, and 200 μg· mL-1 could effectively inhibit the proliferation of NB4 cells, with the cell proliferation inhibition rates of 6. 7 % ,7.4 % ,24. 3 % for 24 hours treatment,2. 3% ,18% ,45.9% for 48 hours and 3.7% ,24% ,51.7% for 72 hours treatment,respectively. Morphology change observation indicated characteristics of cell apoptosis and the DNA agarose electrophoresis showed typical DNA ladder. FGVL could upregulate Bax mRNA and downregulate mRNA level of Bcl-2 in NB4 cells and the effect was positively correlated with the dose and treatment time. Conclusion FGVL could significantly induce apoptosis of NB4 cells, which might be related to the down-regulation of Bct-2 and up-regulation of Bax.
