摘  要:为验证由蚯蚓中提取的蛋白组分—蚓激酶与血栓形成及血栓形成后溶解血栓的关系,本实验采取了动物实验的方法进行药理学研究。首先取雄性大鼠75只,将其随机分为五组,分别命名为空白对照组、阳性药对照组、低剂量组、中剂量组和高剂量组。通过小动物血栓形成仪造模,然后分别对五组给予生理盐水(1.0ml/kg)、低剂量注射液(0.3mg/kg)、中剂量注射液(0.6mg/kg)、高剂量注射液(1.2mg/kg)以及阳性药注射液(1.1ml/kg),给药方法为每日尾静脉给药,连续给药15天。15日后测量血液凝血酶时间(thrombin time TT)、血浆复钙时间(recalcification time,RT)和血小板最大聚集率,并剥离颈部动脉血栓测定血栓重量。结果显示,蚓激酶对于大鼠血液的RT、TT时间均有有效的延长作用,并且对于血栓形成后的血栓溶解也有明显的作用,且这些作用都与蚓激酶剂量成依赖性关系。本实验结论为蚓激酶极可能具有延长血栓形成时间和使外静脉血栓溶解变小的作用。To verify the extracted protein components by earthworms--lumbrokinase,and the impact on thrombosis and thrombolytic with lumbrokinase, the experimental method adopted animal experiment for pharmacology studies. First, taking 30 male rats and dividing them randomly into five groups, named as blank control group, positive control group, low- dose group, middle- dose group and high -dose group. Through the small animal thrombosis instrument modeling, then the five groups were given normal saline (1.0ml/kg), low- dose injection(0.3mg/kg), middle -dose injection (0.6mg/kg), high - dose injection(1.2mg/kg) and positive drug injection ( 1. 1ml/kg). Method of adminis- tration is intravenously administered per day for consecutive 15 days. After 15 days, we measured blood thrombin time (TT), plasma recalcification time (RT) and the maximum platelet aggregation rate and stripped the neck artery throm- bosis and measure the thrombus weight. The results showed that lumbrokinase can effectively extend the RT, TT for the blood of rats, and it also has significant effect on thrombolysis. And these effects are dose - dependent relationship with the lumbrokinase. The experiment concluded that the lumbrokinase is likely to have a role in prolonging thrombus formation time and dissolving the external thrombosis smaller.
