痤疮患者中Toll样受体的表达及其与IL-8和IL-12水平的相关性

摘　　要：目的探讨Toll样受体2（TLR2）在痤疮发病中的作用及其可能的机制。方法收集Ⅰ～Ⅳ级不同严重程度痤疮患者各35例,应用流式细胞术检测其外周血CD14＋单核细胞（PBM）TLR2的表达,同时采用双抗体酶联免疫吸附法（ELISA）检测血清IL-8、IL-12水平。另取35例正常人作为对照组。结果各组痤疮患者外周血单核细胞表面TLR2的表达均显著高于正常人对照组（P〈0.01）,各组患者血清中IL-8和IL-12的水平也显著高于正常人对照组（P〈0.01）。各组患者TLR2的表达与IL-8呈正相关（r分别为0.452、0.583、0.735、0.648,P〈0.05）,与IL-12水平亦呈正相关（r分别为0.365、0.493、0.526、0.461,P〈0.05）。痤疮严重程度与TLR2的表达呈正相关（r=0.325,P〈0.05）,与IL-8水平成呈正相关（r=0.426,P〈0.05）,与IL-12水平也呈正相关性（r=0.627,P〈0.05）。结论 TLR2的表达水平和炎症因子的分泌与痤疮严重程度相关,TLR2的表达在痤疮的发生和发展中起重要作用。Objective To explore the role and possible mechanism of Toll like receptor 2（TLR2）in the pathogenesis of acne.Methods Flow cytometry was performed to detect the expression of TLR2 on peripheral blood CD14＋ monocytes in 35normal controls and four groups of ance patients including 35 cases in each group.according to the severity,levels of IL-8 and IL-12 in sera were also measured by double antibody sandwich ELISA.Results A significant increase was observed in the expression of TLR2 in peripheral blood CD14~＋ monocytes and the level of IL-8 and IL-12 in serum of each patient group compared with the normal controls（P0.01）.The expression of TLR2 was positively correlated with the level of IL-8（r=0.452,0.583,0.735,0.648,P0.05）and IL-12（r=0.365,0.493,0.526,0.461,P0.05）in the four patient groups.The severity degree of acne was positively correlated with the expression of TLR2（r=0.325,P0.05）,the level of IL-8（r=0.426,P0.05） and IL-12（r=0.627,P0.05）.Conclusions The expression of TLR2 and the secretion of IL-8 and IL-12 is related to the severity of acne and the expression of TLR2 plays a critical role in the occurrence and development of ance.
