摘  要:根据虾传染性皮下和造血器官坏死病毒(IHHNV)病毒基因型特点和OIE《水生动物疾病诊断手册》推荐的引物,对一例进境南美白对虾(Litopenaeus vannamei)亲虾进行IHHNV病毒分型PCR扩增并对扩增产物测序,测序结果与NCBI的IHHNV基因序列比对显示同源性为95%以上,确定样品为IHHNV阳性;309F/R引物扩增序列与IHHNV福建分离株、夏威夷分离株、马达加斯加分离株(3A型)、坦桑尼亚分离株(3B型)、泰国分离株进行序列比对,同源性分别为99%、98.7%、79.9%、90.6%和93.9%,显示分离的309bp序列与IHHNV 3A型和3B型的同源性较低,样品为传染性皮下和造血器官坏死病毒感染型毒株I型或Ⅱ型阳性。According to genotypic characteristics of infectious hypodermal and hematopoietic necrosis virus of shrimp and the primers recommended by Manual of Diagnostic Tests for Aquatic Animal 2012 of OIE, virus genotyping of IHHNV from Litopenaeus vannamei was performed by PCR amplification, and the sequence homology reached 95%compared to IHHNV DNA fragment in NCBI. The results demonstrated that this Litopenaeus vannamei was positive IHHNV. The sequence amplified by the specific primers 309F/R from the samples had 99%, 98.7%, 79.9%, 90.6%and 93.9% homology with those from Fujian, Hawaii, Madagascar (3A), Tanzania(3B) and Thailand respectively. T The IHHNV-related sequence of 309bp from the samples showed lower homology compared with Type 3A(from Madagascar)and Type 3B(from Tanzania) ,which are not infectious IHHNV . The results demonstrated that the samples are infectious IHHNV.
