摘  要:目的:研究有氧运动对内皮祖细胞诱导大鼠心肌微血管新生能力的影响。方法:健康2月龄雄性SD大鼠20只,按体重随机分为空白对照组和有氧运动组。采用递增负荷跑台训练建立有氧运动模型,每周训练6 d,为期8周。通过双荧光染色法观察细胞对乙酰化低密度脂蛋白的摄取和与荆豆凝集素1的结合,结合形态学特征鉴定内皮祖细胞,倒置显微镜下计数贴壁细胞数量以反映细胞增殖和黏附情况。结果:1)CD34较好标记了心肌微血管,有氧运动组大鼠心肌微血管内皮上较对照组CD34(P<0.05)和AC133蛋白表达多(P<0.01);2)外周血单个核细胞中CD34+细胞比例有氧运动组更高(P<0.01),CD34+/Flk-1+细胞所占比例2组无差异(P>0.05);3)细胞双荧光染色呈双阳性,表明所培养贴壁细胞为内皮祖细胞;4)第48 h和第7 d的有氧运动组外周血与骨髓来源的内皮祖细胞数量较对照组都多(P<0.05);5)7 d内,对照组外周血和骨髓源贴壁细胞数量均增殖(P<0.01),有氧运动组外周血源贴壁细胞(P<0.05)和有氧运动组的骨髓源贴壁细胞也增殖(P<0.01);6)第10 d的黏附能力检测结果显示,有氧运动组较对照组贴壁细胞数量增加(P<0.01)。结论:有氧运动促进大鼠心肌微血管新生的机制可能是有氧运动促进大鼠骨髓中内皮祖细胞的动员和增殖,从而释放入血,使循环中内皮祖细胞数量增多、黏附能力增强,并归巢于大鼠心肌微血管壁后,参与诱导微血管的新生。Objective: The study focused on effects of aerobics on angiogenesis in cardiac tissue induced by endothelial progenitor cells in rats. Methods: Twenty two-months old male Sprague-Dawley rats were included in this study,and they were divided into a control group and a aerobics group. Aerobics group rats were participated in graded load treadmill training, 6 days/week, 8 weeks in total. The endothelial progenitor cells were confirmed by uptake of acetylated LDL and binding Ulex europaeus agglutinin 1 ( UEA - 1 ) based on double fluorescent staining. Endothelial progenitor cells were identified by morphological characteristics. Number of adherent cells was calculated under inverted microscope to reflect cell proliferation and adhesion. Results: 1 ) CD34 preferably marked microvessel in cardiac muscle. Endothelial CD34 cell and AC133 expression are more/higher in aerobics group than those in control group (P 〈 0.05, P 〈 0.01 ). 2) There are more CD34^ + cells in peripheral blood in aerobics group than in control group ( P 〈 0.01 ), while no difference of CD34 ^+/Flk - 1 ^+ cell quantity in both groups (P 〉 0.05 ). 3 ) Cell performed double positive means that the cultured adherent cell is endothelial progenitor cell. 4) Quantity of endothelial progenitor cells in 48 h aerobics and 7 days aerobics groups is higher than in control group ( P 〉 0.05 ). 5 ) In the seventh day, quantity of adherent cells derived from peripheral and bone marrow in both control and aerobics groups is increased (P 〈 0.01 ). 6) In the tenth day, adhesion ability shows that quantity of adherent cells in aerobics group is larger than in control group (P 〈 0.01 ). Conclusion : The mechanism of that aerobics promotes angiogenesis in cardiac tissue of rats, is that aerobics could promote mobilization and proliferation of endothelial progenitor cells in bone marrow, which released to peripheral blood and increased the quantity of endothelial progenitor cells in blood, and increase t
