摘 要:目的:研究水通道蛋白8(AQP8)在正常妊娠和羊水过少孕妇胎膜和胎盘中的表达,和地塞米松对羊水过少胎盘、胎膜中AQP8的干预效应。方法:选择2009年10月至2010年10月在遵义医学院附属医院产科住院的足月孕妇30例,其中羊水过少组15例,正常对照组15例。采用RT-PCR技术和免疫组织化学法检测两组在羊膜上皮细胞、胎盘合体滋养细胞和绒毛膜滋养细胞中AQP8 mRNA和蛋白的表达,及分别加入不同剂量的地塞米松(0μg/ml、1μg/ml、5μg/ml、10μg/ml、50μg/ml)孵育24小时后,检测两组产妇胎膜和胎盘组织中AQP8 mRNA的表达水平。结果:两组绒毛膜滋养细胞中AQP8 mRNA和蛋白的表达比较,差异均无统计学意义(P>0.05),而羊水过少组中AQP8 mRNA和蛋白在羊膜上皮细胞和胎盘合体滋养细胞的表达低于正常对照组(P<0.05)。两组羊膜上皮细胞和胎盘合体滋养细胞AQP8 mRNA的表达随着地塞米松浓度的增加有增加趋势。两组羊膜上皮细胞和胎盘合体滋养细胞在地塞米松(50μg/ml)作用24小时后,AQP8 mRNA的表达明显增加,与地塞米松0μg/ml浓度比较,差异有统计学意义(P<0.05),而绒毛膜滋养细胞在不同浓度中的表达比较无差异(P>0.05)。结论:羊膜上皮细胞、胎盘合体滋养细胞与羊水量有一定的关系;地塞米松对AQP8 mRNA表达水平有促进作用,地塞米松可能通过上调羊膜上皮细胞、胎盘合体滋养细胞中AQP8的表达而对羊水量的稳态产生一定的影响。Objective:To evaluate the aquaporins(AQPs) expression in fetal membranes and placenta of pregnant women with normal pregnancy or oligohydramnios, and to research the effects of dexamethasone on the expression and localization of AQP8 in oligohydramnios placenta and fetal membranes. Methods:30 women with full-term birth who hospitalized in Accessorial Hospital of Zunyi Medical College from Oct. 2009 to Oct.2010 were enrolled,including 15 cases with oligohydrarnnios and 15 cases with normal pregnancy as control group. Cells of placenta and fetal membrane were cultivated,and cultured with dexamethasone for 24 hours in various doses(0 μg/ml,1μg/ml,5μg/ml,10 μg/ml,and 50 μg/ml). The expression of AQP8 mRNA in the cells of placenta and fetal membranes was examined by RT-PCR. The location of AQP8 protein in placenta and fetal membrane was examined by immunohistochemistry. Results:There was no significant difference in the expression of either AQP8 mRNA or protein in chorion cytotrophoblasts between two groups. The expressions of both AQP8 mRNA and protein in amniotic epithelial cells and syneytiotrophoblasts were significantly lower in oligohydramnios group than those in control group. In both two groups,AQP8 mRNA expression was positively correlated with the increase of dexamethasone concentration in both amniotic epithelial cell and syneytiotrophoblasts. AQP8 mRNA expression increased significantly in amniotic epithelial cells and syneytiotrophoblasts after 24 hours treatment with dexamethasone(50 μg/ml) in comparison with that without dexamethasone treatment( P 〈0. 05)however, no change was observed in chorion cytotrophoblasts among dexaethasone treatment in various doses( P 〉0. 05). Conclusions: Amniotic epithelial cells and syneytiotrophoblasts are involved in the generation of amniotic fluid,and dexamethsone can increase the AQP8 mRNA expression.
