胃上部癌组织中联合检测Pokemon、P53蛋白对判断外科分子切缘的意义

摘　　要：目的在胃上部癌组织中通过联合检测Pokemon及P53蛋白表达情况,判断外科手术切缘相对安全的范围。方法对60例胃上部癌的中心部位以及在人体解剖纵轴方向距离肿瘤边缘起1.0 cm、1.5 cm、2.0 cm、2.5 cm、3.0 cm、3.5 cm、4.0 cm、4.5 cm、5.0 cm处上下分别取材,然后采用Western blot检测每处Pokemon,P53蛋白的表达情况,并进行统计学分析。结果胃癌组织中Pokemon、P53高表达,在癌旁组织中随着距离增加,此两种蛋白表达相应减少。在癌旁组织中P53,Pokemon蛋白的表达同时在2.5 cm处出现统计学差异,在距离3.5 cm及以上组织中的相对表达率为阴性。结论以Pokemon、P53蛋白的表达作为标志,发现3.5 cm及以外的组织中此两种蛋白的表达与正常组织无差异,对外科手术分子水平的切缘距离的确定可能具有一定指导意义。Objective To determine the relative safety of the surgical margin range through combined detection of Pokemon and P53 in the upper gastric cancer tissues. Methods In 60 cases of upper gastric cancer, specimens were get from tumor tissues and tumor edge of 1.0 cm, 1.5 cm, 2.0 cm, 2.5 cm, 3.0 cm, 3.5 cm, 4.0 cm, 4.5 cm, 5.0 cm . Then using Western blot analysis to detect the Pokemon and P53 protein expression, and analyzed statistically. Results High expression of Pokemon, P53 were found in the tumors＇ cental tissue. The farther distance of tumor margin, the lower expression of Pokemon, P53 proteins. Compared with the center of tumor, the expression of the Pokemon, P53 proteins were significant different at the 2.5 cm. When the incisal margin distance 3.5 cm, the positive expression rate of Pokemon,P53 proteins were 0. Conclusion In Pokemon, P53 protein expression as a marker, expression of these two proteins found no difference in the upper gastric tissues at 3.5 cm and outside 3.5 em with normal tissue, which may have guiding to determine the distance to the surgical margins.
