祛银颗粒对角质形成细胞生长因子诱导的HaCaT细胞增殖及分泌TNF-α的影响

摘　　要：目的探寻中药组方祛银颗粒治疗银屑病的作用机制。方法体外培养HaCaT细胞,以10 ng/mL的KGF刺激HaCaT细胞增殖的方法建立银屑病的细胞模型。用祛银颗粒不同浓度含药血清干预细胞,倒置显微镜下观察药物作用后的细胞生长状态,ELISA法检测细胞分泌TNF-α的影响,流式细胞仪检测药物血清对细胞增殖、细胞周期的影响。结果 48 h祛银颗粒低、中、高剂量组药物与模型组比较,对细胞均有抑制作用(P<0.05);祛银颗粒中、高剂量组抑制作用与维A酸组差异无统计学意义(P>0.05)。流式细胞仪检测显示:在细胞合成S期,祛银颗粒高剂量组同期细胞数量较模型组减少(P<0.05),与维A酸组相近(P>0.05)。祛银颗粒高剂量组与模型组比较,分泌的TNF-α降低(P<0.05)。结论祛银颗粒含药血清具有抑制KGF诱导的HaCaT细胞增殖的作用,并可抑制细胞分泌TNF-α,提示抑制表皮增殖和抑制细胞产生炎症因子可能是祛银颗粒发挥治疗银屑病的作用机制之一。Objective：To explore the mechanism of Quyin granula treating psoriasis.Methods：HaCaT cells in vitro culture proliferation induced by 10 ng/ml of KGF was used to establish cell model of psoriasis.Cell growth after medication was observed under inverted microscope.ELISA method was used to detect TNF-α secreted by cell supernatant.Flow cytometry instrument was applied to detect the effect of drug serum on cell proliferation and cell cycle.Results：Compared with model group,Quyin granula low,medium,and high dose groups have inhibitory effect on cells （P＜0.05） at 48 hours;the inhibitory effect of Quyin granula in high dose group and Tretinion group were of no statistical difference （P＞ 0.05）.Flow cytometry test showed：during the synthesis of cells in S phase,cell number of Quyin granula high-dose group decreased compared with model group,and the difference was statistically significant（P＜ 0.05）,but compared with Tretinion group,there was no significant difference （P＞ 0.05）.Quyin granula high dose group reduced to secrete TNF-α compared with model group （P＜ 0.05）.Conclusions：Quyin granula serum can inhibit the proliferation of HaCaT cells induced by KGF and affect the secretion of TNF-α.
