摘 要:目的观察当归补血汤对人循环内皮祖细胞(EPCs)功能及B细胞淋巴瘤/白血病-2基因(Bcl-2)表达的影响。方法分离培养人外周血单个核细胞,7 d后对贴壁细胞行EPCs鉴定后,将EPCs随机分为6组:空白组、对照组、辛伐他汀组、当归补血汤高、中、低浓度组。空白组用不含血清的培养液培养;对照组用O.2%的牛血清白蛋白培养;辛伐他汀组用0.1μmol/L的辛伐他汀培养;当归补血汤高、中、低浓度组分别用浓度4、2、1 g/L的当归补血汤培养。各组培养24 h后移去上清,加入不含血清的培养液继续培养24 h。采用四唑盐(MTT)法检测细胞增殖力,黏附实验检测黏附力,迁移小室检测迁移力,体外血管生成试剂盒检测成小管力,RT-PCR法检测Bcl-2 mRNA表达,蛋白免疫印迹法检测Bcl-2蛋白的表达。结果当归补血汤高浓度组及辛伐他汀组可显著促进EPCs的各项功能并上调细胞Bcl-2 mRNA的表达(P<0.05);当归补血汤中、低浓度组对EPCs的增殖、迁移及成小管功能具有促进作用,但对细胞黏附力和Bcl-2 mRNA表达无明显影响;当归补血汤高、中、低浓度组及辛伐他汀组均可上调EPCsBcl-2蛋白的表达(P<0.05)。结论当归补血汤可通过上调Bcl-2的表达来促进循环EPCs的功能。Objective To observe the influences of Danggui Buxue Tang on function of circulating endothelial progenitor cells (EPCs) and Bcl-2 expression. Methods Human mononuclear ceils in peripheral blood were isolated and cultured for 7 d. After adherent cells identified, EPCs were randomly divided into 6 groups: blank group, control group, simvastatin group, and high-dose, mid-dose and low- dose Danggui Buxue Tang groups (high-dose group, mid-dose group and low-dose group). Blank group was cultured with nutrient solution without medicated serum, control group, 0. 2% bovine serum albumin, simvastatin group, 0.1 ~mol/L simvastatin, and high-dose group, mid-dose group and lowdose group, respectively, 4 g, 2 g and 1 g Danggui Buxue Tang. The supermatant was eliminated 24 h after culturing, and then all groups were given nutrient solution without medicated serum for continuous culturing for 24 h. The proliferation ability, adhesive force, migration ability and tubule formation of EPCs were detected, respectively, by using MTT, adhesion test, Transwell assay and in vitro angiogenesis kit. The expressions of Bcl-2 mRNA and Bcl-2 protein were detected respectively by using RT-PCR and Western blot. Results All functions of EPCs were significantly improved and expression of Bcl-2 mRNA was up-regulated in high-dose group and simvastatin group (P 〈 0.05 ). The proliferation,migration of and tubule formation of EPCs were improved but adhesive force and expression of Bcl-2 mRNA were not affected in mid-dose group and low-dose group. The expression of Bcl-2 protein was up-regulated in high-dose group, mid-dose group, low-dose group Conclusion Danggui Buxue Tang can improve the function expressions of Bcl-2. and simvastatin group ( P 〈 0.05 ). of EPCs through up-regulating the
