摘 要:目的探讨三七总皂苷对脑梗死后骨髓间充质干细胞动员的作用。方法参照改良ZeaLonga法建立大鼠大脑中动脉脑缺血再灌注损伤模型(MCAO)。造模成功大鼠随机分为模型组、三七总皂苷高、中、低剂量组。实验各组再随机按1、7、14、28 d时间点分为亚组。三七总皂苷高、中、低剂量组分别以60、40、20g/L浓度的血塞通胶囊粉末灌胃,每天10 mL/kg,正常组和模型组以等体积生理盐水灌胃,1次/d,共28 d。流式细胞仪检测不同时间点血液和骨髓中CD54、CD106、CD105、CD117阳性细胞百分数,ELISA法检测不同时间点血液和骨髓中干细胞因子(SCF)表达情况。结果三七总皂苷高、中剂量组和模型组的外周血和骨髓中SCF、CD117、CD105阳性细胞百分数从第1天开始升高,14 d到高峰,其后逐渐降低,但三七总皂苷高、中剂量组上升趋势优于模型组(P<0.05);模型组、三七总皂苷低、中、高剂量组外周血和骨髓中CD54、CD106在第1天到高峰,其后逐渐降低。三七总皂苷中、高剂量组与模型组相比各时间点表达均降低,其中三七总皂苷高剂量组降低明显(P<0.05)。结论三七总皂苷通过促进SCF和降低CD54、CD106表达,促进骨髓间充质干细胞从骨髓动员到外周血中,提高归巢干细胞数量。Objective To investigate the mobilizing effect of panax notoginseng saponins (PNS) on marrow mesenchymal stem cells after cerebral infarction. Methods The rat model of MCAO was established by referencing Zea-Longa method. The modeled rats were randomly divided into model group and high-dose, mid-dose and low-dose Xuesaitong Capsules groups (high-dose group, mid-dose group and low-dose group) , and then according to time points divided into l-d, 7-d, 14-d and 28-d subgroups. High-dose group, mid-dose group and low-dose group were treated with Xuesaitong Capsules (60 g/L, 40 g/L and 20 g/L, 10 g/kg once a day) , and normal group and model group were orally given normal saline water once a day for 28 d. The positive percentage of CD54, CD106, CD105 and CDl17 in peripheral blood and bone marrow were detected by using flow cytometer at different time points, and expression of stem cell factor (SCF) in peripheral blood and bone marrow was detected by using ELISAat different time points. Results SCF level and positive percentage CD117 and CD105 increased in high-dose group, mid-dose group and model group on the first day, reached the peak on the 14th day, and then decreased gradually, but the increased trend is superior in high-dose group and mid-dose group to model group (P 〈 0.05 ). The expressions of CD54 and CD106 reached the peak in model group, high-dose group, mid-dose group and model group on the first day and then decreased gradually, and decreased in mid-dose group and high-dose group at all time points compared with model group especially in high-dose group ( P 〈 0.05 ). Conclusion PNS can mobilize marrow mesonchymal stem cells from bone marrow to peripheral blood and promote number of homing stem cells through improve SCF level and decreasing expressions of CD54 and CD106.
