复方何首乌饮对5/6肾切除大鼠肾保护作用的研究

摘　　要：目的探讨复方何首乌饮对5/6肾切除大鼠慢性肾衰竭模型的肾脏保护作用及其可能的机制。方法大鼠行5/6肾切除术制造动物模型,造模成功后给药,于给药前及给药后4、8、12周采集样本,测定24 h尿蛋白、血尿素氮、血清肌酐的量及转化生长因子-β1(TGF-β1)mRNA在肾的表达。结果复方何首乌饮能显著性地降低24 h尿蛋白排泄量(P<0.05),降低血尿素氮、血清肌酐含量(P<0.05),同时抑制大鼠肾TGF-β1 mRNA的表达(P<0.05)。结论复方何首乌饮对5/6肾切除大鼠肾具有保护作用,其可能的机制为抑制TGF-β1 mRNA的表达。Objective To investigate the protective effect of compound recipe Heshouwuyin on kidney of chronic renal failure （ CRF ） rats and its possible mechanism .Methods The rats were 5/6 nephrectomized to establish animal models,after the models were successfully established , the rats were administrated .The specimens were collected before medication and 4,8,12 weeks after administration,then 24h urine protein quantity,BUN,Scr were detected,in additional,the expression levels of TGF-β1 mRNA in kidney were determined .Results The compound recipe Heshouwuyin could significantly decrease 24h urine protein quantity,the levels of Scr and BUN （ P 〈0.05）,meanwhile,which could inhibit the expression of TGF-β1 mRNA in nephridial tissue （ P 〈0.05 ）.Conclusion The compound recipe Heshouwuyin has protective effect on kidney of CRF rats , and the action mechanism may be related with its inhibitory effect on expression of TGF-β1 mRNA.
