摘 要:目的探讨百草枯一次性腹腔注射致小鼠肺纤维化的病理改变,进而筛选出病理改变稳定的小鼠肺纤维化模型。方法昆明(KM)小鼠、ICR小鼠及C57BL/6J小鼠各36只,分别分为模型组及对照组。模型组一次性小鼠腹腔注射百草枯溶液10 mg/kg或20 mg/kg,对照组注射相同体积的生理盐水。所有小鼠均于建模后第28天处死,比较观察三个品系小鼠肺组织羟脯氨酸含量,HE染色观察肺组织的病理学改变,Masson染色观察肺组织胶原纤维的改变。结果①随着给药时间的延长,三组小鼠活动力均逐渐下降,被毛杂乱,呼吸急促,消瘦并出陆续现死亡,其中C57BL/6J小鼠体重下降最明显,KM小鼠体重稍下降后复升。②三个品系小鼠模型组与对照组相比羟脯氨酸含量差异均有统计学意义(P<0.001),其中KM小鼠10mg/kg组及20 mg/kg组差异无统计学意义(P>0.05),而C57BL/6J模型组小鼠羟脯氨酸含量较其余两个品系增高(P<0.01),ICR小鼠与KM小鼠模型组间差异无统计学意义(P>0.05)。③光学显微镜下观察KM小鼠及ICR小鼠肺组织均无明显纤维化改变,仅C57BL/6J小鼠部分区域有轻度纤维化形成。结论与KM小鼠及ICR小鼠相比,C57BL/6J小鼠更适合构建小鼠肺纤维化模型。Objective To explore a better animal model of pulmonary fibrosis. Methods KM, ICR and C57BIM6J mice were divided into model group and control group ,respectively. The mice received intrapefitoneal injection of 10 mg/kg or 20 mg/kg parquat(PQ) and normal saline, respectively. All mice were sacrificed at 28 d. Part of left lung tissues were used for detecting the contents of hydroxyproline. HE and Masson stains were used to evaluate the degree of pulmonary fibrosis in mice. Results①Aftcr giving the parquat, the activity of all mice started to decrease, and all mice showed hair messy, weight loss, shortness of breath, and some of mice died. The C57BIM6J mice had the most obvious weight loss, while the body weight of KM mice first decreased and then returned to the normal. ② Compared to control group, the hydroxyproline contents in the three strains were all increased in model groups ( P 〈 0. 001 ). Hydroxyproline contents of C57BL/6J model mice were higher than those of the other two strains, but the hydroxyproline contents were not siginificantly different between ICR and KM model mice. ③No fibrosis was seen in ICR and KM mice. Only a small part of pulmonary fibrosis was found in C57BIM6J mice. Conclusion The C57BIM6J mice is more appropriate for establishing the models of pulmonary fibrosis than KM and ICR mice by intraperitoneal injection of parquat.
