摘  要:目的研究高血磷对大鼠肾脏Ⅱ型及Ⅲ型钠磷(NaPi)协同转运子基因表达的影响。方法将48只SD大鼠,分成3组:高血磷组(HP)注射果糖二磷酸钠;低血磷组(LP)喂食自制低磷饲料;正常血磷组(NP)喂食正常饲料,分别于第1、2、4、6周处死3组中各4只样本,以3-磷酸甘油醛脱氢酶(GAPDH)为参照基因,用半定量逆转录-聚合酶链反应(RT-PCR)检测NaPi-Ⅱa mRNA和NaPi-ⅢmRNA在大鼠肾脏的表达情况,同时测定血清钙(Ca),磷(P)及甲状旁腺激素(iPTH)。结果 (1)第1、2、4、6周HP组较LP组和NP组血P及iPTH均明显增高(P<0.05),3组各阶段血清Ca差异无统计学意义(P>0.05);(2)HP组较LP组和NP组肾脏NaPi-Ⅱa mRNA表达明显减少(P<0.05),且HP组随着时间的增加NaPi-Ⅱa表达减少(P<0.05);(3)LP组NaPi-Ⅱa较NP组在第4周开始明显增加(P<0.05);(4)第1、2、4周HP组与LP组、NP组比较,NaPi-ⅢmRNA表达差异无统计学意义(P>0.05),第6周HP组NaPi-ⅢmRNA表达比LP组和NP组高,且差异有统计学意义(P<0.05)。LP组与NP组比较,NaPi-ⅢmRNA表达差异无统计学意义(P>0.05)。结论高血磷显著抑制NaPi-Ⅱa转运体mRNA的表达和影响NaPi-Ⅲ转运体mRNA的表达,同时高磷血症是促进大鼠iPTH增高的、不受钙影响的独立因素。Objective To investigated the effect of hyperphosphatemia on the expression of rat renal typeⅡ and typeⅢ sodium phosphorus(Na-Pi)cotransporter gene (Na-PiⅡa and Na-PⅢI).Methods 48 SD rats were divided into three groups:the first group was the hyperphosphatemia group (HP)and injected with fructose diphosphate sodium,the second group was the hypophos-phatemia group(LP)and fed with the self-made low phosphorus fodder and the third group was the normal blood phosphorus group (NP)and fed with normal fodder.4 rats in each group were killed at 1,2,4,6 weeks respectively.With 3-phosphoglyceraldehyde dehydrogenase(GAPDH)as the reference gene,the semi-quantitative retrovirus-polymerase chain reaction (RT-PCR)test was ap-plied to detect the expression of NaPi-Ⅱa-mRNA and NaPi-Ⅲ-mRNA in the rat kidney.At the same time serum calcium (Ca2+), phosphorus (P)and intact parathyroid hormone (iPTH)were detected.Results (1)The serum P and and iPTH at 1,2,4,6 weeks in the HP group were significantly higher than those in the LP group and the NP group (P〈0.05);the serum Ca2+ in the HP,LP and NP groups had no obvious differences in various periods(P〉0.05);(2)the expression of renal Na-PiⅡa mRNA in the HP group was significantly decreased compared with the LP and NP groups,moreover,with the time increase,the expression of Na-PiⅡa mRNA in the HP group was reduced (P〈0.05);(3)compared with the NP group,the Na-PiⅡa mRNA expression in the LP group began to significantly increase at 4 weeks(P〈0.05);(4)compared with the LP and NP groups,the expression of Na-PiⅢ mRNA at 1,2,4 weeks in the HP group had no obvious difference (P〉0.05),while the Na-PiⅢ mRNA expression at 6 weeks in the HP group was higher than that in the LP and NP groups with statistical differences (P〈0.05).The Na-PiⅢ mRNA expres-sion in the LP and NP groups had no significantly difference(P〉0.05).Conclusion Hyperphosphatemia significantly inhibit the exp
