摘 要:目的:探讨新生小鼠鼠巨细胞病毒(MCMV)肝炎治疗前后外周血及肝脏中 Th17相关性细胞因子白细胞介素-17(IL-17)、白细胞介素-22(IL-22)的表达变化及其意义。方法腹腔接种 MCMV 建立 MCMV 肝炎模型(病毒组),对照组腹腔注射等量的无菌生理盐水,更昔洛韦治疗组在接种病毒后24 h 开始每天腹腔注射更昔洛韦,连用2周。分别用 ELISA 方法和 RT-PCR 检测小鼠外周血中 IL-17、IL-22水平和肝组织 IL-17 mRNA、IL-22 mRNA 的表达。结果病毒组小鼠血清 IL-17、肝组织 IL-17 mRNA 在感染后第3天开始增高,且呈持续升高趋势,直至第14天表达最高,与对照组比较差异有统计学意义(P 〈0.01);治疗7、14 d 后血清 IL-17、肝组织 IL-17 mRNA 水平较病毒组相应时点下降(P 〈0.01)。病毒组小鼠血清 IL-22、肝组织 IL-22 mRNA 在感染后第3天开始增高,第7天达高峰,第14天有所下降,与对照组比较差异有统计学意义(P 〈0.01);治疗14 d 后血清 IL-22、肝组织 IL-22 mRNA 较病毒组相应时点增高(P 〈0.01)。结论 IL-17可能参与了新生小鼠 MCMV 肝炎的发病,IL-22在 MCMV 肝炎中起保护作用。Objective To investigate the variation of interleukin-17( IL-17)and interleukin-22 (IL-22)in peripheral blood and liver before and after therapy in newborn mice with MCMV hepatitis. Methods BALB / c mice were infected by intraperitoneal injection of MCMV(Virus group),the mice in control group were given intraperitoneal injection of the same amount of sterile saline,and the mice in treatment group were injected with ganciclovir of MCMV daily for 2 weeks 24 hours after inoculation. The periperal blood and liver levels of IL-17,IL-22 were evaluated by ELISA and semi-RT-PCR. Results The serum levels of IL-17 protein, liver levels of IL-17 mRNA increased in the MCMV mice from the 3th day to the 14th day. The serum levels of IL-22,liver levels of IL-22 mRNA in-creased in the MCMV mice from the 3th day to the 14th day,and the levels reached to the peak on the 7th day,and then IL-22 slightly decreased on the 14th day. The differences were significant compared with the control group(P 〈 0. 01). The serum levels of IL-17,liver levels of IL-17mRNA decreased in the treatment group from the 7th day to the 14th day. Conclusions IL-17 maybe involved in the patho-genesis of MCMV hepatitis and IL-22 play a protective role in the MCMV infection.
