摘 要:本研究采用PCR—RFLP方法对苏太、梅山、大白、杜洛克和长白猪CD14基因-61位点多态性进行分析,同时应用ELISA方法检测苏太、梅山猪的IL-2、IL-6、IL-10和IL-12水平,分析苏太、梅山猪CD14基因多态性对部分免疫指标的遗传效应.探讨将该位点作为遗传标记运用于猪抗病育种的可行性。结果表明:苏太、梅山和大白猪CD14基因-61位点经BamH I酶切后,可检测到AA、AG、GG3种基因型,而杜洛克和长白猪中只检测到AG和GG2种基因型:苏太、梅山猪与3个引进品种的基因型分布均表现为差异极显著(P〈0.01),大白猪与长白猪的基因型分布均表现为差异不显著(P〉0.05):遗传多态性分析表明苏太猪、梅山猪的PIC值介于0.25—0.5,表现为中度多态.大白、杜洛克和长白猪的PIC值低于0.25,表现为低度多态;品种效应对所测免疫指标影响极显著(P〈0.01),且在苏太猪和梅山猪中.不同基因型间各免疫指标水平差异均不显著(P〉0.05)。因此,CD14基因-61位点的突变不会对苏太、梅山猪的一般抗病力产生显著影响,有必要继续筛选CD14基因的功能突变位点和一般抗病力的有效遗传标记.为下一步抗病育种工作提供指导和依据。The gene polymorphism at -61 locus of CD14 gene in Sutai, Meishan, Large White, Duroc and Landrace were investigated by PCR-RFLP method, the level of IL-2, IL-6, IL-10 and IL-12 of Sutai and Meishan pigs were detected by ELISA method, and analyzed the genetic effect of CD14 gene polymorphism on some immune indices in Sutai and Meishan pigs, which aimed at providing feasibility analysis of this polymorphic site for further disease-resistant breeding of pigs. The results showed that -61 locus of CD14 gene of Sutai, Meishan and Large White digested by BamH I could be divided into three kinds of genotypes, AA, AG and GG, while only two genotypes, AG and GG were detected in Duroc and Landrace. The difference of genotype distribution was extremely significant among Sutai, Meishan and three foreign pig breeds (P〈0.01), and there was no significant difference among Large White and Landrace (P〉0.05). The analysis of genetic polymorphisms indicated that PIC value of Sutai and Meishan was range from 0.25 to 0.5, which displayed moderate polymorphism, while PIC value of Large White, Duroc and Landrace was less than 0.25, which displayed low polymorphism. The measured immune indices were extremely significantly influenced by effect of varieties in Sutai and Meishan (P〈0.01), while the difference of immune indices between different genotypes was not significant. The mutation at -61 locus of CD14 gene would not make significant impact on the general disease resistance of Sutai and Meishan pigs, which indicated that it is necessary to continue to probe functional mutation sites and effective genetic markers of CD14 gene, and then provide guidance and theoretical foundation for further disease-resistant breeding of pigs.
