摘　　要：目的用高效阴离子交换色谱-脉冲安培检测器联用技术建立苦瓜多糖组分的分析方法。方法以苦瓜为原料,用水提醇沉法提取苦瓜粗多糖,并对其工艺进行探讨。苦瓜粗多糖经超声波酸水解处理后,以12 mmol/L NaOH溶液为淋洗液,流动相流速为1.0mL/min,CarboPaC PA10(250mm×4mm i.d.)为分析柱,进样量为25μL进行分析测定。结果苦瓜多糖中主要含有阿拉伯糖、果糖、葡萄糖、甘露糖和半乳糖,各种单糖组分在一定浓度范围内线性关系良好,能很好地分离及准确测定。结论该方法操作简便,灵敏度高,重现性好。Objective To developed a method of quantitative determination of momordica polysaccharide components by high performance anion exchange chromatography coupled with pulsed amperometric detection.Methods The optimized extraction conditions of momordica polysaccharide were tested with water extraction and alcohol precipitation method by orthogonal technique.Using acid hydrosis by ultrasonic treatment for the crude polysaccharide,the separation was performed on a CarboPaC PA10（250mm × 4mm I.D.） anion exchange column with 12mmol/L NaOH solution as the eluent at a fl ow rate of 1.0mL/min.The injection volume was 25 μL.Results The main contains of momordica polysaccharide were arabinose,fructose,glucose,mannose and galactose.All kinds of monosaccharide composition showed good liner relationship under the optimum conditions.The detection limits（S/N=3）were 1.8μg/L,5.3μg/L,2.8μg/L,3.8μg/L,2.5μg/L,and the average recoveries were 96.5%,102.0%,97.5%,102.5%,99.0% with good reproducibility for arabinose,fructose,glucose,mannose and galactose,respectively.Conclusion This method has the advantages of simple operation,high sensitivity and good reproducibility.
