逍遥散提取物对瘦素诱导活化人肝星状细胞的影响

摘　　要：目的探讨逍遥散提取物对瘦素诱导人肝星状细胞活化相关细胞因子及细胞外基质表达的影响。方法以瘦素刺激体外培养人肝星状细胞(HSC-LX2)活化,用逍遥散提取物进行干预;24 h后,免疫细胞化学法检测α-SMA和TGF-β1蛋白表达,ELISA法检测COL-Ⅰ、COL-III和HA的含量。结果与空白对照组比较,瘦素刺激组α-SMA和TGF-β1的蛋白表达以及COL-Ⅰ、COL-III和HA的含量均明显增加(P<0.05,P<0.01)。与瘦素刺激组比较,逍遥散组α-SMA和TGF-β1的蛋白表达以及COL-Ⅰ、COL-III和HA的含量均明显降低(P<0.05,P<0.01)。结论逍遥散提取物抗肝纤维化的作用机制可能与调节肝细胞中α-SMA和TGF-β1蛋白的异常表达,降低胶原含量有关。Objective To explore the effects of ＂Xiaoyao Powder＂ extract on expressions of related cell factors and extracellular matrix in hepatic stellate cells induced by leptin. Methods Leptin was used to stimulate HSC-LX2 activating, and ＂Xiaoyao Powder＂ extract was used for the intervention. Expressions of α- SMA and TGF-β1 were detected by immunocytochemical method and the contents of COL- I , COL-Ill and HA were detected by ELISA after 24 hours. Results Compared with blank control group, the expressions of α-SMA and TGF-I31 , and contents of COL- I , COL-Ⅲ and HA were increased in leptin stimulating group （P 〈0.05, P 〈 0.01 ）. Compared with leptin stimulating group, the expressions of α-SMA and TGF-β1, and contents of COL- 1 , COL-β1 and HA were decreased markedly （ P 〈 0.05, P 〈 0.01 ）. Conclusion The mechanism of ＂Xiaoyao Powder＂ extract may be association with regulating the abnormal protein expressions of α-SMA and TGF-β1 and reducing collagen content to anti-liver fibrosis.
