摘  要:目的 建立异基因大鼠联体共生的动物模型 ,探讨其诱导肾移植耐受机理。 方法 对DA和LEW大鼠实施联体手术并诱导耐受 ,然后进行DA→LEW的肾移植。FACS检测术后脾脏、胸腺中嵌合状态 ;检测体内、外过继转移耐受大鼠脾细胞对正常大鼠单向混合淋巴细胞反应 (MLR)和迟发性过敏反应 (DTH)的抑制作用 ,以及T细胞克隆不应答在耐受中的作用。 结果 联体共生诱导耐受可使DA→LEW的肾移植存活明显延长。耐受组脾脏、胸腺中供体细胞的嵌合比例明显高于排斥组。耐受可通过体内、外过继转移给正常大鼠。外源性IL 2可部分逆转耐受状态。 结论 嵌合状态与肾移植耐受存在明显相关性 ,克隆不应答。Objective To establish rat models for studying the relation between chimerism and transplantation tolerance and to study the mechanism of renal allograft tolerance induced by parabiosis combined with cyclophosphamide(Cp) on rats. Methods Renal transplantation tolerance was induced by parabiosis combined with Cp ,and the renal transplantation was performed on DA→LEW rats. Chimeritic cells of donor origin in recipient spleen and thymus were detected by FACS after transplantation. Suppressive role of tolerant spleen cells adoptively transferred in vivo and in vitro was tested by DTH and MLR. Meanwhile the role of T cells clonal anergy was also detected. Results Parabiosis combined with Cp prolonged the survival of DA→LEW rat renal allografts obviously. Chimeritic status in spleen and thymus of toleranced rats was obviously higher than that in rejected rats,and this status could be adoptively transferred in vivo and in vitro, which can also be reversed partly by exogenous IL 2. Conclusions There is close correlation between chimeritic status and renal transplantation tolerance. Several other mechanisms may by theorized for the induction of tolerance in this renal transplant model, including clonal anergy and suppressive cells.
