摘 要:结合国家标准GB/T15686—2008与实验室的实际情况,建立高粱中单宁含量的测定方法。对显色反应时间、酸碱性、溶液反应温度等方面进行了反复的验证。经过对单宁标准系列的测定,结果表明,显色剂与单宁反应后溶液颜色变化显著,用分光光度计在波长为525 nm处测定,单宁质量浓度在0.1-5 mg/mL范围内服从朗伯-比耳定律,回归方程为A=0.9640c+0.219 3,相关系数R2为0.995 4。经过对17个高粱品种样品单宁含量进行测定,该方法的回收率在97.3%-103.4%范围内。该方法简便快捷、灵敏度高、特异性好、结果准确,适合大批量样品分析,可应用于高粱中单宁含量的检测。In this paper, the method for determination of tannin content in sorghum was set based on the national standards GB/T15686-2008 and the actual situation in the laboratory. Chromogenic reaction time, acidity and basicity, solution temperature and other items are repeatedly verified. After a series of determination, the results showed the color of solution changes significantly after the reaction of chromogenic reagent and tannin. Detecting at the wave-length of 525 nm with a spectrophotometer, we found tannin concentration obeyed Lambert-Beer law within the range from 0.1 mg/mL to 5 mg/mL. The regression equation was A=0.964 0c+0.219 3 and R2=0.995 4. After the determination of tannin content in samples of 17 sorghum varieties, we found the recovery was 97.3%-103.4%. With features like simplicity, rapidity, high sensitivity, good specificity and accuracy, this method is suitable for large-volume sample analysis and could be used to detect the content of tannin in sorghum.
