摘  要:以甜柿与涩柿叶片为试材,采用3种不同有机溶剂作为提取液,比较研究了甜柿和涩柿2种柿树叶片叶绿素含量的测定方法.结果表明:采用研磨法提取叶绿素时,溶剂以体积分数为95%的乙醇最好,甜柿叶片叶绿素总含量、叶绿素a含量和叶绿素b含量分别为1.428、1.086和0.341mg/g,涩柿叶片叶绿素总含量、叶绿素a含量和叶绿素b含量分别为0.951、0.737和0.213mg/g,均显著高于体积分数为80%的丙酮和二者的混合液;采用浸泡法提取叶绿素时,以体积分数为80%的丙酮和体积分数为95%的乙醇按体积比1:1混合的混合液提取效果最好,甜柿叶片叶绿素总含量、叶绿素a含量和叶绿素b含量分别为1.478、1.089和0.388 mg/g,涩柿叶片叶绿素总含量、叶绿素a含量和叶绿素b含量分别为1.180、0.886和0.294 mg/g,均显著高于体积分数为95%的乙醇和体积分数为80%的丙酮.两种叶绿素提取方法比较发现,浸泡法优于研磨法.With sweet persimmon and astringent persimmon leaves as experimental material,the effects of three extraction solvents and two extraction methods on chlorophyll content were studied.The results showed that,when the chlorophyII was extracted by grinding method,the extraction efficiencies by using 95% ethanol alone were better than mixed solvent or 80% acetone alone.The content of total chlorophyll,chlorophyll a,chlorophyll b in sweet persimmon leaves was 1.428 mg/g,1.086 mg/g,and 0.341 mg/g respectively;The content of total chlorophyll,chlorophyll a, chlorophyll b in astringent persimmon leaves was 0.951 mg/g,0.737 mg/g,and 0.213 mg/g respectively.When the chlorophyll was extracted by soaking method,the extraction efficiencies by using mixed solvent were better than 95% ethanol or 80% acetone alone.The content of total chlorophyll,chlorophyll a,chlorophyll b in sweet persimmon leaves was 1.478 mg/g,l.089 mg/g and 0.388 mg/g respectively;The content of total chlorophyll,chlorophyll a,chlorophyll b in astringent persimmon leaves was 1.180 mg/g,0.886 mg/g,and 0.294 mg/g respectively.When the extraction efficiencies by grinding method was compared with that by soaking method,the latter was better.
