摘 要:目的:测定金莲花中金丝桃苷的含量。方法:采用80%乙醇提取总黄酮,HPLC法测定金莲花不同部位中金丝桃苷的含量。色谱柱为Phenomenex Gemini C18柱(4.6mm×250mm,5μm);流动相为乙腈-0.05%磷酸溶液(V/V,18:82);流速为1.0ml/min;柱温为25℃;检测波长为360nm;进样量为10μl。结果:金丝桃苷在0.04。0.56μg范围内与峰面积呈良好的线性关系,相关系数为0.9997(n=6);其中茎、叶、籽中金丝桃苷的含量分别为0.0279,0.3868,0.0016mg/g。结论:金莲花不同部位中金丝桃苷的含量分布为:叶>茎>籽,上述结果可为金莲花的综合利用提供科学依据。Objective: To determine the content of Hyperoside in Trollius chinensis. Methods: 80% ethanol was used to extracted total flavonoids. HPLC assay was used to determine the Hyperoside content in different parts of Trollius chinensis. The column was Phenomenex Gemini C18 column (4.6 mm×250 ram,5μm);mobile phase of acetonitrile-0.05% phosphoric acid(V/V, 18:82) ;flow rate of 1.0 ml/min;The column temperature was 25 ℃, and the deteetion wavelength was 360 nm,and the injection volume was 10 μl. Result: Within the range of 0.04~0.56 μg, Hyperoside showed a good linear correlation with peak area with the coefficient of 0.99969 (n=6);Hyperoside contents in the stems,leaves, and seeds were 0.0279 mg/g, 0.3868 mg/g, 0.0016 mg/g respectively. Conclusion: The distribution of Hyperoside contents in different parts of Trollius chinensis is as follows: leaf〉stem〉seed. The above results provide scientific basis for comprehensive utilization of Trollius chinensis.
