摘  要:目的 检验Cdk5 / p35在神经元成熟和萌芽中起着重要作用的假说。 方法 采用Westernblot实验、免疫沉降和Cdk5活性分析的方法 ,研究出生后各阶段大鼠三叉神经节中该激酶的活性变化和Cdk5、p35的表达水平。结果 随着大鼠三叉神经节的发育 ,Cdk5的活性逐渐增加。新生大鼠三叉神经节中的Cdk5活性比成体正常大鼠三叉神经节中的Cdk5活性高约六倍。这一激酶活性的变化与 p35表达的变化是一致的。Cdk5的表达量在各阶段中是恒定不变的。结论 结果提示了Cdk5 / p35与神经元的分化。Objective To test the hypothesis that Cdk/p35 plays important roles in neuronal maturation and sprouting.Methods We studied the changes of Cdk5 activity,expression levels of Cdk5 and p35 in the development rat trigiminal neurons by Western blot methods、immunoprecipitation and kinase assay.Results By degrees the expression of p35 significantly decreased from newborn to adult rat trigiminal neurons.The expression of p35 was prominent in newborn rat trigiminal neurons,and it was most lowest in adult rat trigiminal neurons.In contrast the expression of Cdk5 was constant throughout the development rat trigiminal neurons.Cdk5 activity in the newborn rat trigiminal neurons was about 6 times the height of that in normal adult rats trigiminal neurons.The changes of the kinase activity coincided well with those of p35 expression in the development rat trigiminal neurons.Conclusion These data suggest that Cdk5/p35 plays important roles in synaptic reorganization,and expression of p35 may be a novel regulatory mechanism to control Cdk5 activity.
