摘  要:目的:比较来自不同生态环境、不同地理位置及不同形态和不同生理生化特性的六株钝项螺旋藻的系统进化关系和遗传多样性,试图从DNA水平探讨它们长期在不同的生存环境作用下遗传进化关系和种质资源的多样性,为螺旋藻种间系统发育关系的研究及正确地进行种间分类提供参考。方法:用RAPD分子标记技术和ITS序列对六株不同来源螺旋藻进行扩增,利用PopGen32、MEGA 5.0等软件分析其遗传多样性,构建系统发育树并进行进化地位的比较。结果:RAPD结果显示,不同来源的六株螺旋藻分为两大枝,FACHB-HY独为一枝与其他五株亲缘关系最远。Spi-wz、FACHB-350、FACHB-793、FACHB-904和FACHB-834聚为一枝,而FACHB-834与其他四株的关系较远,Spi-wz和FACHB-904、FACHB-793和FACHB-350亲缘关系最近,ITS基因的同源性比较结果与RAPD分子标记的结果一致。结论:来自不同生态环境的螺旋藻株在长期的进化地位和系统发育关系上存在有差异,表现出遗传多样性和种资资源的多样性。RAPD分子标记和ITS序列同源性聚类结果相一致。Objective: To compare the phylogenetic relationships and genetic diversity of six kinds of Spirulina platensis which come from different ecological environments, different locations and different morphology and different physiological and biochemical characteristics. The genetic evolutionary relationships and the diversity of germplasm resources from the level of DNA were studied and then a reference for the phylogenetic research and interspecific classified correctly of Spirulina platensis were provided. Methods: The technique of RAPD molecular marker and ITS, PopGen32, MEGA 5 and other software were used to analyze the genetic diversity and construct phylogenetic tree and their evolutionary status was compared. Results: RAPD showed that the six kinds of Spirulina platensis were divided into two branches, FACHB-HY had a furthest kinship with the others. Spi-wz, FACHB-350, FACHB-793, FACHB-904 and FACHB-834 clustered into one branches, while FACHB-834 and the other four distantly related, Spi-wz and FACHB-904, FACHB-793 and FACHB- 350 had a closest relationship. The homology comparison of ITS are consistent with the result of RAPD. Conclusion: Spirulina platensis from different habitats exist differences in the long-term evolution status and phylogenetic relationships, showing the diversity of the genetic and germplasm resources. The homology comparison of ITS is consistent with the result of RAPD.
