摘 要:目的:动态观察单侧输尿管结扎(UUO)大鼠肾组织中α-平滑肌肌动蛋白(α-SMA)、E-钙黏蛋白(E-cadherin)的表达及益肾化瘀方对其影响。方法:将清洁级雄性SD大鼠108只随机分为假手术组,模型组,益肾化瘀方高、中、低剂量组及缬沙坦组,除假手术组外各组采用UUO模型,分别于术后第7、14、28天光镜下观察肾间质病理改变,免疫组化法观察大鼠肾组织α-SMA、E-cadherin的表达变化。结果:造模后7d,大鼠肾组织中α-SMA表达即明显增高,而E-cadherin表达逐渐减少;与模型组相比,各治疗组大鼠α-SMA的表达减弱,E-cadherin的表达增强(P<0.01,P<0.05),以益肾化瘀方高剂量组和缬沙坦组变化最显著,二者比较无统计学意义。结论:益肾化瘀方可能通过抑制大鼠肾组织中α-SMA的早期高表达,上调E-cadherin的表达,而发挥抗肾间质纤维化作用。Objective: To observe the effect of Yishen Huayu Recipe on the expression of α-SMA and E-cadherin on renal tissues of unilateral ureteral obstruction (UUO) rats. Methods: 108 clean SD male rats were randomly divided into shamoperation group, model group, high, medium and low dose of Yishen Huayu group, Valsartan group. Except the sham-operation group, the other groups were using the UUO model. Qualitative pathological changes of renal interstitial were observed under light microscope on the postoperative day 7, 14 and 18. Immunohistochemistry method was used to observe the expression changes of α-SMA, E-cadherin on rat renal tissue. Results: On the 7th day after establishment of UUO model, the expression of α-SMA on renal tissues of rats increased significantly, while the expression of E-cadherin reduced gradually. Compared with the model group, the expression of α-SMA in the treatment groups decreased while the expression of E-cadherin increased significant (P〈0.01, P〈0.05), especially the changes in the high dose of Yishen Huayu group and valsartan group, but there was no statistical significance between the two groups. Conclusion: Yishen Huayu Recipe can resist renal interstitial fibrosis possibly through decreasing early high expression of α-SMA and increase the expression of E-cadherin in renal tissues of rats.
