益气消癥方对慢性阻塞性肺疾病模型大鼠MMP-9和TIMP-1表达的影响

摘　　要：目的:观察益气消癥方对慢性柱塞性肺疾病(COPD)模型大鼠基质金属蛋白酶抑制剂-1(MMP-9)和基质金属蛋白酶抑制剂-1(TIMP-1)表达的影响,探讨其防治COPD的作用机制。方法:30只大鼠随机分为正常组、模型组和益气消癥组,每组10只。采用香烟烟雾暴露联合气道内注入脂多糖(LPS)法制备COPD模型。实验第2天起,模型组、益气消癥组分别给予0.9%氯化钠溶液、益气消癥方灌胃。实验第113天处死各组大鼠,用免疫组化法检测气道上皮MMP-9、TIMP-1的表达。结果:模型组气道上皮MMP-9、TIMP-1表达较正常组升高,MMP-9/TIMP-1比值较正常组降低,差异均具有统计学意义(P<0.05);益气消癥组TIMP-1表达较模型组显著降低(P<0.05)。结论:益气消癥方能够通过降低COPD大鼠气道上皮MMP-9、TIMP-1表达,调节MMP-9/TIMP-1平衡来防治COPD,其中以降低TIMP-1表达、改善气道重构作用更为明显。Objective： To observe the effect of Yiqi Xiaozheng Recipe on the expression of MMP-9 and TIMP-1 in rats with Chronic Obstructive Pulmonary Disease （COPD）, and investigate the mechanism of preventing and treating COPD. Methods： 30 rats were randomly divided into normal group, model group and Yiqi Xiaozheng group, with 10 rats in each group. The rat model of COPD was established by passive smoking and intratracheal instillation of lipopolysaccharide （LPS）, except the normal group. From the 2nd day, the model group and the Yiqi Xiaozheng group were given 0.9% sodium chloride and Yiqi Xiaozheng recipe respectively. On the 113rd day, all the rats were sacrificed, and the expression of MMP-9 and TIMP-1 were detected by immunohistochemistry method. Results： Compared with the normal group, the expressions of MMP-9 and TIMP-lof model group increased while the MMP-9/TIMP-1 ratio decreased, the differences were statistically significant （P〈0.05）; Compared with model group, the expressions of MMP-9 and TIMP-lof Yiqi Xiaozheng group decreased while the MMP-9/TIMP-1 ratio increased, the difference of the expression of TIMP-1 was statistically significant （P〈0.05）. Conclusion： Yiqi Xiaozheng recipe can prevent and treat COPD by reducing the expression of MMP-9 and TIMP-lon airway epithelium in rat with COPD, regulating the MMP-9/ TIMP-1 ratio balance, of which decreasing the expression of TIMP-1 and the effect of improving airway remodeling were more obvious.
