摘  要:目的:探讨具有益气养阴活血解毒作用的降糖舒心方对内质网应激凋亡通路的影响及逆转糖尿病心肌重构作用。方法:30只MKR小鼠随机分为MKR模型组,降糖舒心方组,GB组(格列喹酮+贝那普利组),每组10只,另取10只野生型C57鼠作对照。给药前、给药30d后各检测空腹血糖(FBG)1次,实验结束后测量小鼠血清甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)等,心肌细胞凋亡用TUNEL法检测;内质网应激信号通路分子糖调节蛋白78(GRP78)和C/EBP同源蛋白(CHOP),用免疫组化检测蛋白表达水平,用RT-PCR检测mRNA转录水平。结果:与MKR模型组相比,降糖舒心方组能明显降低空腹血糖(FBG)、LDL-C和TG(P<0.05,P<0.01),显著升高HDL-C(P<0.01);显著下调GRP78、CHOP蛋白和mRNA的表达(P<0.01),显著减少心肌细胞凋亡水平(P<0.05)和减轻心肌纤维化(P<0.01)。结论:降糖舒心方具有降糖调脂作用,对内质网应激介导的心肌细胞凋亡具有保护作用,能改善心肌纤维化,逆转心室重塑。Objective: To investigate the effect of Jiangtang Shuxin Formula (JTSXF), which can supplement qi, nourish yin, activate blood and remove toxic agents, on myocardial remodeling caused by endoplasmic reticulum stress-related cells apoptosis in diabetic rats. Methods: 30 MKR rats were randomly divided into 3 groups: MKR model group, JTSXF group and western medicine (Gliquidone+Benazepril, GB) group. Negative control group was the wild-type C57 rats. The fasting blood- glucose (FBG) level was determined respectively before and after the treatment of 30 days. Serum lipid TG, LDL-C and HDL-C were tested after the experiment. Myocardial cell apoptosis was examined by TUNEL. The immunohistochemistry and RT-PCR were respectively used to test the protein and mRNA expression of endoplasmic reticulum stress signaling pathway molecules GRP78 and CHOP. Masson staining was used to test the myocardial collagen fiber. Results: Compared with MKR model group, JTSXF group could significantly decrease the serum lipid level of FBG, LDL-C and TG (P〈0.05, P〈0.01), and significantly increase the lipid HDL-C (P〈0.01), the protein and mRNA expression of GRP78, CHOP, myocardial apoptosis and alleviate myocardial fibrosis (P〈0.01). Conclusion: JTSXF can decrease blood glucose and improve the dyslipidemia, possess a protective effect on endoplasmic reticulum stress-related myocardial cells apoptosis, ameliorate myocardial fibrosis and reverse ventricular remodeling.
