摘 要:目的:观察不同浓度鹿茸提取液在正常和低钙条件下对离体蟾蜍心肌收缩力和心率的影响。方法:60只蟾蜍随机分为任氏液组对照组、任氏液+鹿茸组和低钙任氏液+鹿茸组,采用斯氏法制备离体蛙心灌流标本。在任氏液或低钙任氏液中加入鹿茸提取液,通过蟾蜍心室肌肉收缩波动,心室外加药心室内吸收的方法给药。应用BL-420F生物机能实验系统记录的离体蟾蜍心肌收缩曲线,并进行心肌收缩力和心率的统计学分析。结果:任氏液+鹿茸组:在心室外浓度达到40 mg·mL-1后,心肌收缩力在加药前后比较,心肌收缩力明显增加,P<0.01。低钙任氏液+鹿茸组:在心室外浓度达到5 mg·mL-1后,心肌收缩力在加药前后比较,心肌收缩力明显增加,P<0.05。正常蟾蜍离体蛙心灌流标本的心率、心室外加入鹿茸前后的心率和在低钙条件下心室外加入鹿茸前后的心率均没有显著变化,P>0.05。结论:在低钙条件下,鹿茸提取液对离体蟾蜍心脏活动的兴奋作用比在正常任氏液条件下更容易出现,但对心率没有影响。Objective : To observe the effects of different concentrations of pilose antler extract under normal and low calcium concentration conditions on isolated toad heart contractility and heart rate. Methods : 60 toads were randomly divided into Ringer solution control group, group of Ringer solution+pilose antler extract group and low calcium Ringer solution+pilose antler extract group. The perfusion specimens of isolated frog heart were prepared by Si's method. The ventricle muscle contraction wave of toad was recorded by the outer pilose antler extract administration with inner ventricular absorption way. We recorded and observed the contractile wave curve of isolated toad heart for 30 rain after administration with pilose antler extract. And then myocardial contractility and heart rate were analyzed statistically by BL-420F biological and functional experimental system. Result : The ringer solution+pilose antler group: myocardial contractile extent was increased significantly when the 40 mg · mL^-1 pilose antler extract was added in comparison with that before ( P〈0.01 ). However, low calcium Ringer solution+pilose antler group: when 5 mg· mL-1 pilose antler extract was added, myocardial contractility was increased significantly in comparison with that before (P〈0.01). The heart rate in toad heart ventricular perfusion specimens under normal Ringer solution add pilose antler or low calcium condition had no significant change comparing with that of before ( P〉0.05 ). Conclusion : It was more easy to show the effects of pilose antler extract on isolated toad heart contractile extent in low calcium condition than that of in the normal Ringer solution conditions, however, there was no effect on heart rate.
