摘  要:主要研究了人凝血酶、速乐涓、邦亭、苏灵、巴曲亭对纤维蛋白原的水解作用。通过还原性SDS-PAGE法对作用结果进行检测,并用透射电子显微镜观察各样品在不同时间形成的凝块结构。速乐涓、苏灵、巴曲亭只作用于纤维蛋白原的α链,对β、γ链无作用;邦亭起初作用于纤维蛋白原的β链,对α链作用较弱,随着时间的延长对α链增强,对γ链无作用。相同反应条件下,类凝血酶制剂作用纤维蛋白原的初速度:巴曲亭≥人凝血酶>速乐涓>邦亭>苏灵;反应过程的速度:邦亭>人凝血酶>苏灵>巴曲亭≥速乐涓;凝块纤维直径:巴曲亭>苏灵>速乐涓≥邦亭≥人凝血酶。不同蛇毒来源类凝血酶制剂对纤维蛋白原作用情况有差异,为临床应用提供参考。The hydrolysis of human thrombin,Su Lejuan,Bang Ting,Su Ling and Ba Quting to the fibrinogen to fibrin was mainly studied.The actional results were detected by reduced SDS-PAGE and the fibrin network structures of each sample at different intervals during the clots formation were observed under transmission electron microscope.Basis of the fibers shape and size,the coagulated extent was assessed.Su Lejuan,Su Ling and Ba Quing affected specifically on α-chain of the fibrinogen only,but had no influence on β-chain and γ-chain.However the Bang Ting affected the β-chain of fibrinogen initially,the action of the α-chain was weak,but with prolonging the time,the action of the α-chain enhanced,at the same time showed no effect on the γ-chain.Comparing the initial speed,Baquting≥ human thrombin Su Lejuan Bang Ting Su Ling,but the speed of reaction process was distinct,Bang Ting human thrombin Su Ling Ba Quting ≥Su Lejuan.Comparing the diameter of fibers:Ba Quting≥Su Ling Su Lejuan Bang Ting≥human thrombin.The action of differnet Snake venom thrombin-like enzyme preparations to fibrinogen was dissimilar.
