金边蚂蟥抗痛风作用研究

摘　　要：目的研究金边蚂蟥Poecilobdella manillensis抗痛风作用及安全性。方法小鼠ip次黄嘌呤复制高尿酸血症模型,观察金边蚂蟥对高尿酸血症小鼠及正常小鼠血清尿酸水平的影响;大鼠足跖sc微晶型尿酸钠结晶(MSU),复制痛风性炎症模型,观察金边蚂蟥对痛风性炎症的影响;热板法、扭体法观察金边蚂蟥的镇痛作用;并测定金边蚂蟥对小鼠的急性毒性。结果与模型组相比,金边蚂蟥1.50 g/kg组可以显著降低高尿酸血症小鼠的血液尿酸水平,金边蚂蟥3.00、1.50 g/kg组显著降低正常小鼠血液尿酸水平(P<0.01);金边蚂蟥0.75 g/kg组可显著抑制MSU引起大鼠足跖肿胀(P<0.05、0.01);金边蚂蟥3.00、0.75 g/kg组可显著减少由醋酸引起的小鼠扭体反应次数(P<0.05、0.01);小鼠ig金边蚂蟥的急性毒性试验未观察到明显毒性反应,最大耐受量为生药23.09 g/kg。结论金边蚂蟥具有良好的抗痛风作用。Objective To observe the therapeutic effects on gout and the acute toxicity ofPoecilobdella manillensis. Methods The anti-gout effects ofP. manillensis were studied by observing its effects on the hyperuricacidemia model and the level of blood uric acid. And we observed the effect ofP. manillensis on gouty inflammation via injection of MSU to the foot of rats. The analgesic effect and toxicity of P. manillensis will be reported in this work too. Results Comparing with the model group, P. manillensis could significantly reduce the level of the blood uric acid in the hyperuricemic and normal mice at 3.00 and 1.50 g/kg doses （P 〈 0.01）, and acute gouty arthritis at 0.75 g/kg dose （P 〈 0.05, 0.01）. It reduced the number of writhing in mice at 3.00 and 0.75 g/kg doses （P 〈 0.05, 0.01）. Obvious toxic reaction was not observed and the maximum tolerant dose was 23.09 g/kg for mice in acute toxicity experiment. Conclusion P. manillensis is proved to have the therapeutic effect on gout. Further research and development will be continued.
