摘  要:目的评价发酵培育的蝙蝠蛾拟青霉菌丝体(PHM)抗小鼠力竭疲劳作用及其作用机制。方法采用轮式疲劳仪强迫小鼠每日30 min连续18 d进行攀跑运动,攀跑速度从10.2 m·min-1逐渐递增至12.6 m·min-1,并于第19天(d 19)强迫小鼠攀跑60 min以建立力竭疲劳模型。每日训练前40 min分别ig给予PHM 140,280及560 mg·kg-1和莫达非尼13 mg·kg-1。分别于d5,d9,d13,d17和d19记录小鼠的电击次数,每日记录小鼠的体质量。检测肌糖原(MG)、肝糖原(LG)、血清尿素氮(SUN)、血清乳酸(SLA)和血清肌酸激酶(CK)的含量。结果模型组小鼠在d19力竭训练中的电击次数显著高于给药组,且与正常对照组小鼠相比,其MG及LG含量显著降低,而SUN及CK水平显著升高(P<0.05)。PHM280及560 mg·kg-1组小鼠在力竭训练中的电击次数分别为133±55和115±41次,均显著低于模型组(240±89)次(P<0.01)。PHM 560 mg·kg-1组小鼠的MG和LG分别为1.28±0.24和(40.9±10.2)mg·g-1,均显著高于模型组〔MG:(0.91±0.26)mg·g-1,LG:(22.2±2.9)mg·g-1;P<0.01〕,而SUN,SLA和CK分别为(5.8±1.3)mmol·L-1,(5.7±1.7)mmol·L-1和(0.6±0.2)kU·L-1,均显著低于模型组〔SUN:(7.5±2.1)mmol·L-1,SLA:(7.1±1.8)mmol·L-1,CK:(0.8±0.3)kU·L-1;P<0.05〕。与模型组小鼠相比,莫达非尼组小鼠的电击次数显著降低(P<0.01),但MG,LG,SUN,SLA和CK水平无显著性差异。结论 PHM可能通过增加能源物质的储备,增强机体有氧代谢能力,并减少代谢产物的积累,以维持机体内环境稳定及正常代谢功能,从而起到抗力竭疲劳的作用。OBJECTIVE To evaluate the effect and mechanism of Paecilomyces hepiali mycelium( PHM) on exhausted fatigue in mice. METHODS Mice were forced to run 30 min a day for consecutive 18 d using the wheel running fatigue apparatus, and the speed was gradually increased from 10. 2 m·min^-1to 12. 6 m·min^-1. On d19,these animals were forced to run 60 min to establish an exhausted fatigue model. Every day,40 min before the run training,the animals were ig administered with PHM( 140,280,560 mg·kg^-1) and modafinil( 13 mg·kg^-1). The number of electric shocks was recorded on d5,d9,d13,d17 and d19,respectively,and the body mass of each mouse was recorded daily. The levels of muscle glycogen( MG),liver glycogen( LG),serum urea nitrogen( SUN),serum lactic acid( SLA) and serum creatine kinase( CK) were detected by commercially available kits. RESULTS During the last exhausted running on d19,the number of electric shocks of the model group was markedly larger than that of the drug administration groups. When compared to the normal control group,the model group showed a significant decrease in the level of either MG or LG,but a significant increase in the level of either SUN or CK. The number of electric shocks of the PHM in the moderate and high dose groups was 133 ± 55 and 115 ± 41 times,respectively,which was significantly smaller than that of the model group 240 ± 89( P〈0. 01). The levels of MG and LG of the PHM 560 mg·kg^-1group were( 1. 28 ± 0. 24) mg·g^-1and( 40. 9 ± 10. 2) mg·g^-1,respectively,which were significantly higher than those of the model group 〔MG:( 0. 91 ± 0. 26) mg·g-1,LG:( 22.2 ±2.9) mg·g-1; P〈0.01〕,and the levels of SUN,SLA and CK of this PHM group were( 5. 8 ± 1. 3) mmol·L^-1,( 5.7 ±1.7) mmol·L^-1and( 0.6 ±0.2) kU·L^-1,respectively,which were significantly lower than those of the model group〔SUN:( 7.5 ±2.1) mmol·L^-1,SLA:( 7.1 ±1.8) mmol·L^-1,CK:( 0.8 ±0.3) kU·L^-1; P〈0.05〕.
