摘 要:目的探讨体外CSC/SBA-15固化材料对人骨髓间充质干细胞(hMSCs)功能的影响。方法将有序介孔材料SBA-15与硫酸钙骨水泥(CSC)粉末以5%、10%和20%质量比复合制备CSC/SBA固化材料,分为单纯CSC组(CSC组)、5%质量比组(CSC-5S组)、10%质量比组(CSC-10S组)和20%质量比组(CSC-20S组)。四组材料与hMSCs培养12h后,采用MTT测定细胞黏附,1、3、7d后采用MTT法测定细胞增殖。与成骨诱导液共培养,7、14d后进行碱性磷酸酶(ALP)定量,21d后茜素红染色分析。结果与细胞共培养12h后,CSC-10S及CSC-20S组材料表面黏附细胞数量大于CSC及CSC-5S组(P<0.05)。7d时,CSC-5S组、CSC-10S组和CSC-20S组的细胞增殖率高于CSC组(P<0.05),CSC-10S组和CSC-20S组表面细胞的ALP活性高于CSC组和CSC-5S组(P<0.05)。14d时,随着SBA-15含量增加,CSC/SBA-15材料的ALP活性定量水平逐渐提高;21d时,CSC-5S、CSC-10S及CSC-20S组材料表面矿化结节的茜素红染色增强。结论 CSC/SBA-15固化材料有利于hMSCs的黏附、增殖及向成骨细胞分化,有望应用于骨缺损的修复。Objective To investigate the in vitro effect of CSC/SBA-15 materials on the function of human mesenchymal stem cells (hMSCs). Methods The ordered mesoporous material SBA-15 was blended into calcium sulfate cement(CSC) powder with 5M, 10% and 20M mass ratio to make CSC/SBA-15 compound materials, which was devided into 4 groups of CSC(pure CSC cement), CSC-5S(5 % mass ratio),CSC-10S(10 % mass ratio) and CSC-20S(20 % mass ratio). The samples of 4 groups were cultured with hMSCs. After cultured for 12 hours, the cell adherence on the material surface was determined by MTT. The cell proliferation was determined by MTT on the 1st, 3ra and 7th day after culture. The hMSCs attached on the samples were cultured with osteoinductive a-MEM medium. The alkaline phosphatase(ALP) quantitative analysis was performed on the 7th and 14th day and alizarin red staining analysis was performed on the 21st day. Results After cultured for 12 hours, the cell number adhered on the CSC-10S and CSC-20S surface was obviously higher than that on the CSC and CSC-5S surface(P〈0. 05). After cultured for 7 days, the cell proliferation rate in groups of CSC-5S,CSC-10S and CSC-20S was significantly higher than that in group CSC(P〈0. 05). Compared with groups of CSC and CSC-5S after 7 days culture, the ALP activity in groups of CSC-10S and CSC-20S was obviously increased (P〈0. 05). As the SBA-15 mass ratio increased, the ALP activity in CSC/SBA-15 was increased on the 14th day(P〈0. 05). Alizarin red staining of mineralized nodule on the 21st day was more obvious in groups of CSC-5S, CSC-10S and CSC-20S than that in group CSC. Conclusion Compared with pure CSC, compound CSC/SBA-15 material is more prone to hMSCs adhesion,proliferation and osteoblast differentiation, and hopefully to be used in bone defect repairing.
