摘  要:目的探讨孟鲁司特对脓毒症大鼠肺组织半胱氨酰白三烯受体1(CysLT1R)表达的影响。方法将48只SD大鼠随机均分成模型组及孟鲁司特治疗组。用盲肠结扎穿孔及导管留置法制备脓毒症急性肺损伤大鼠模型。治疗组术前30min用孟鲁司特钠咀嚼片按10mg·kg-1·d-1剂量稀释后给大鼠灌胃,持续至实验结束。两组分别在术后6、12、24、48h分别处死6只大鼠,取肺组织;用免疫组化法检测肺组织CysLT1R的表达,RT-PCR法检测肺组织CysLT1RmRNA表达,并观察肺组织的病理变化。结果各时间点治疗组大鼠肺组织CysLT1RmRNA和CysLT1R表达均低于模型组(P<0.05)。治疗组肺组织病理损伤较模型组减轻(P<0.05)。结论孟鲁司特能下调脓毒症大鼠肺组织CysLT1R的表达,对脓毒症大鼠肺损伤起到保护作用。Objective To study the influence of montelukast on cysteinyl leukotriene receptor-1 (CysLTIR) expression in the lungs of rat model with sepsis. Methods The sepsis model with acute lung injury was established by cecal ligation and puncture in 48 SD rats, which were equally assigned into two groups. With gastric gavage,the rats in group A were treated with montelukast 10 mg · kg-1 · d-1 at 30 rain before operation. The rats in group B were taken as model controls. At 6,12,24 and 48 hours,6 rats from each group were sacrificed at each time point for detecting the expression of CysLTIR in the lungs by immunohistochemistry and RT-PCR The pathological changes in the lungs were observed as well. Results The expressions of CysLT1R mRNA at all time points were lower in group A than those in group B (P〈0. 05 ). So did the expressions of CysLT1R (P〈0. 05). The pathological injury of lung tissues was less group A than that in group B (P〈0. 05 ). Conclusion Montelukast has a protective effect on the sepsis-induced acute lung injury in rats by down-regulating the expression of CysLT1R in the lungs.
