摘　　要：建立了间接 ELISA定量测定生长激素释放因子 ( GRF)的方法。其基本程序为 ,抗原包被、5%牛血清白蛋白和 5%猪血清封闭、一抗反应 (山羊抗人 GRF1～ 2 9抗体 )、二抗反应 (生物素化兔抗山羊 Ig G)、链霉卵白素 -HRP反应、DAB显色。最佳工作条件为 ,一抗 1∶ 1 6 0 0、二抗 1∶ 32 0 0、链霉卵白素 -HRP0 .5mg/L。在上述条件下 ,绘制了标准曲线 ( 2～ 4 0 ng) ,并测定了中国仓鼠肾细胞 ( CHO)表达A new ELISA method was developed to detect Growth Hormone Releasing Factor(GRF) in this study. The ELISA procedure included the encapsulation of antigen, first antibody reaction(goat anti-hGRF 1-29), second antibody reaction(biotinylted rabbit anti-goat IgG), streptavidin HRP reaction and DAB reaction. The optimized conditions for ELISA were: first antibody 1∶1 600, second antibody 1∶3 200, streptavidin HRP 0.5 mg/L. A standard curve was drawn ranging from 2 ng to 40 ng of GRF. This experiment detected GRF concentration in transfected CHO supernatant successfully with this method.
